[Objective]To detect metallothioneinâ… /â…¡(MTâ… /â…¡) and cytochrome c (Cyt C) inhippocampus of pentylenetetrazol (PTZ)-induced epilepsy young rats.And to explorethe effects of mouse nerve growth factor (mNGF) on expression of MTâ… /â…¡and CytC)in hippocampus of pentylenetetrazol (PTZ)-induced epilepsy young rats.[Methods]Fifty19-day-old young male SD rats were randomly divided into fivegroups(groupA~E). Each group had10rats.Group B was injected withpentylenetetrazol (PTZ)40mg kg-1.Group C~E were respectively injected withmNGF500AU kg-1,1000AU kg-1,2000AU kg-1after PTZ40mg kg-1d-1.Group A were injected with equal dose of saline. Real-time PCR was used to detectMT â… and Cyt C mRNA in the hippocampus of rats.Andimmunohistochemistry wasused to detect MT â… /â…¡and Cyt C positive cells in the hippocampus of rats.[Results]1. Weight and behavior: comparing with the negative control group, theweight,diet, activity reduced significantly in each group with PTZ-induced epilepsy.And the weight,diet, activity of group C~E gradually increased after given low,medium, after high dose mNGF.2. The MTâ… mRNA expression of group B was significantly higher than group A(P <0.01).The MTâ… mRNA expression of group C~E after given low, medium andhigh dose mNGF were less than group B (P <0.0001). The MTâ… mRNA expression ofgroup E was significantly less than group C (P <0.0001).3. The Cyt C mRNA expression of group B was significantly higher than groupA (P <0.01).The Cyt C mRNA expression of group C~E after given low, mediumand high dose mNGF were less than group B (P <0.0001). The Cyt C mRNAexpression of group E and D was significantly less than group C (P <0.001).4. Immunohistochemistry was used to detect MTâ… /â…¡positive cells, and theMTâ… /â…¡ positive cells of Group B was significantly greater than group A (P <0.01);the MTâ… /â…¡ positive cells of group C~E after given low, medium and high dosemNGF were less than group B (P <0.01); the MTâ… /â…¡ positive cells of Group C was significantly greater than D, E group (P <0.0001).5. Immunohistochemistry was used to detect Cyt C positive cells, and the Cyt Cpositive cells of Group B was significantly greater than group A (P <0.01); the Cyt Cpositive cells of group C~E after given low, medium and high dose mNGF were lessthan group B (P <0.01); the Cyt C positive cells of Group C was significantly greaterthan D, E group (P <0.0001).[Conclusions]1. The expression of MTâ… and Cyt C increased in pentylenetetrazol(PTZ)-induced epilepsy young rats.2. mNGF may have a protective effect for brain tissue with epilepsy. |
