Epilepsy On The 1st Of Pentylenetetrazol Induced Epilepsy Model In Rat Hippocampus Apoptosis Regulation Factor Of P53, The Expression Of Fas And Caspase-3 Expression And Its Clinical Efficacy Observation

Objective: In this study, we investigated Epilepsy Formula No. 1(EF.1) in the rats with epilepsy and p53,fas,caspase-3 protein expression in hippocampus of epileptic model rats. To make a preliminary assessment about the curative effect of Epilepsy Formula No.1. And we also investigated the probalble mechanism of action.Methods: 60 Male and 60 Female SD rats, weighed 200 ~250g. The model of epileptic paroxysm was set up. 100 severe ones were filted from the rats with epilepsy according to the grade of EF.1. Then the rats were divided into 6 groups according to the factorial design: Model group,Chinese medicine Epilepsy Formula No.1 (low, middle, high dose)group,western medicine group,normal group. Each group has 20 rats. Model group,Chinese medicine Epilepsy Formula No.1 (low, middle, high dose)group and western medicine group were all injected with 32mg/kg pentylenetetrazol (PTZ) by intraperitoneal injection, once a day for 3 weeks. After 7 days without PTZ, they were all injected by the same ways and same dose. Then Chinese medicine Epilepsy Formula No.1 groups were treated respectively with 0.4g/kg, 0.8g/kg, 1.6g/kg EF.1 by gastric perfusion daily. The western medicine group was received 43mg/kg carbamazepine by gastric perfusion daily. The model group was received the same volum Sodium Chloride by gastric perfusion daily. After treating for 4 weeks, the materials of hippocampus could be drawn from the rats and be investigated by immunohistochemistry. And we could investigated the Apoptosis cells in hippocampus by microscope.Results: 1. During the process of gastric administration, the mortality in the 5 groups were not statistically significant.(P>0.05)2.The break-out times and break-out grade in Chinese medicine(EF.1)groups and western medicine group became fewer and lower(P<0.05) .There were no significant difference between them (P>0.05) .3.The number of p53,fas,caspase-3 protein positive expression in hippocampus of model group were obviously more than the normal group(P<0.05) . And to be compared with Chinese medicine Epilepsy Formula No. 1 and western medicine group, p53,fas,caspase-3 protein positive expression in hippocampus were obviously less than the model group(P<0.05) .In the same time, There were no significant difference between the highest dose of EF.1 groups and group (P>0.05) .Conclusions: Epilepsy Formula No.1 can step down the population of cell apoptosis, and have better anti-apoptotic action .The anti-epileptic effective maybe associated with the inhibiting p53,fas,caspase-3 protein expression in model rats' hippocampus.
