| Musa spp. cv. Fenjiao is one of fine banana cultivars, however, their multiplication coefficient of rapid propagation is low. In this experiment, Musa spp., cv. Bendifenjiao (AAB. Group from Fuzhou) was used as the materials, the effects of solid and liquid modes and different LED light sources on proliferation and rooting were analysed; ISSR molecular marker was also employed for analysis the differentiation of genetic stability between liquid and solid culture modes; meanwhile, the economic benefits analysis between solid and liquid modes were also performed. This study would provide technical supporting for industrializing propagation.The main results were showed as follows:1. The effects of solid and liquid culture on micropropagationâ‘ The establishment of sterile system. Firstly, explants were sterilized with 75% ethanol for 40s, then transfered into 0.1% HgCl2 solution for 15 min. The initiation medium was MS+0.4 mg/L 6-BA+0.4mg/L NAA+40g/L sugar+6 g/L agar, the germination rate was 42.3%.â‘¡ The optimization of solid proliferation medium. Different levels of 6-BA and sugar were choosen as the test factors, completely random experiments were carried out to optimizate the proliferation medium, the results indicated that the optimal solid proliferation medium was MS +0.1 mg/L NAA+3.0 mg/L 6-BA+6 g/L agar agar+30 g/L sugar, the proliferation rate was 2.02.â‘¢ The optimization of liquid proliferation model. The effects of container type, density of fluid volume, inoculation quantity,6-BA concentration, rotate speed on the proliferation in the liquid cultured medium were studied. Results indicated that, the optimal conditions were using 250 ml conical flask as container, with 13 mL of liquid medium supplement with 3.0 mg/L 6-BA, best inoculated quantity was 3 plantlets per bottle, seedlings were cultured with a shaking table under 80 r/min rotation speed, and the proliferation rate was up to 5.13.â‘£ The optimazation of rooting liquid cultured mode. Effects of 4 NAA concentrations (0.2 mg/L,0.3 mg/L,0.4 mg/L,0.5 mg/L) on 6 indexes including rooting rate, root length, seedling height, root number, stem height, stem diameter were analysis. Radar comprehensive analysis was employed to evaluate the results. After these, that optimal liquid rooting medium was 1/2 MS+1 g/L AC+0.4 mg/L NAA+30 g/L sugar, the rooting rate was 100%.2. The Effects of different LED light sources on micropropagationâ‘ Effects of different LED light sources on micropropogation. The plantlets were cultured on MS+0.1mg/LNAA+3.0 mg/L 6-BA+6g/L Agar+30g/L solid medium,6 color of LED light sources including red, blue, yellow, green, white and warm-white were choosen for evaluating the effects of different LED lignt on micropropogation, fluorescent lamps were used as contol. Results indicated that, those dwarf banana placed under 5 color LED light sources including red, blue, green, white and warm-white were showed lower proliferation coffients than those placed under fluorescent lamps(control), meanwhile, those placed under red and blue LED lights showed higher proliferation cofflents, and their proliferation coefficients were 2.88 and 2.89, respectively.â‘¡ Effects of different LED light source on rooting. The plantlets were cultured on 1/2MS +0.4 mg/L NAA+1g/L AC rooting medium,6 color of LED light sources including red, blue, yellow, green, white and warm-white were choosen for evaluating the effects of different LED lignt on rooting, fluorescent lamps were used as contol. Indexes including rooting rate, root length, seedling height, root number, stem height, stem diameter were analysed. Results indicated that blue LED lights would suitable for rooting and strengthening, and their rooting rates were upto 100%.3. Analysis of genetic stability between the plantlets cultured on liquid and solid modes by ISSR molecular markerTotal DNA were extracted form leave of dwarf banana cultured on liquid and solid medium, 11 UBC primers showed high polymorphism were choosen for PCR amplification. Results indicated that, total 90 bands were amplified, the gene variation between those cultured on liquid and solid medium were different, the variation rate of plantlets cultured on liquid medium was 4.49%, the the variation rate of plantlets cultured on solid medium was 3.37%. After all, there were not significant differentiations between 2 cultured models. NTSYS software analysis indcated that, plantlets cultured on liquid and solid media showed high genetic stability within 9 generations.4. The economic benefit comparison analysis of micropropagation between solid-and liquid cultures.â‘ Economic benefit analysis of micropropagation in solid cultured mode. The solid medium was MS+0.1 mg/L NAA+3.0 mg/L 6-BA+6 g/L agar+30 g/L sugar, after 25 d, the multiplication coefficient was 2.02; the optimal solid rooting medium was 1/2 ms+0.5 mg/L IBA +40 g/L sugar+0.4 mg/L NAA+1.0 g/L AC+6 g/L agar, after 25 d, rooting rate was 100%.1 million plantlets after transfer by solid cultutre mode totally costed 327550 yuan RMB. The process of micropropagation accounted for the largest cost of total investment, was 62.54%, transplanting fees accounted for 30.20%, production equipment depreciation cost and loss cost accounted for 5.73%. The total income of 1 million seedlings was 600000 yuan, the profit of 1 million plantlets using solid culture was 272450 yuan.â‘¡ Economic benefit analysis o fmicropropagation in liquid cultured mode. The proliferation medium was MS+0.1mg/LNAA+2.0mg/L6-BA+30 g/L sugar, after 15 d, the multiplication coefficient was 1.52; the optimal liquid rooting medium was 1/2MS+1g/L AC+0.4mg/L NAA +30g/L suger, after 25 d, rooting rate was 100%.1 million seedlings after transfer in liquid cultured mode totally costed 297652 yuan, lower than in solid cultured mode. The process of micropropagation accounted for the largest cost of total investment, was 54.07%, the transplanting fees account for 33.24%, production equipment depreciation cost and loss cost accounted for 11.01%. he total income of 1 million seedlings was 600000 yuan, the profit of 1 million plantlets using solid culture was 29898 yuan.â‘¢ Comparation economic benefit of the solid-liquid mode for producting 1 million plants, the profie in liquid cultured mode was 29898 yuan higher than in solid cultured mode. Moreover, to producted 1 million seedlings in solid cultured mode took about 1 year, and in liquid cultured mode taked about half a year. Comprehensive analysis on the time cost and the economic benefit, liquid cultured mode for micropropogation was superior to the solid cultured mode. |
