Lignan compounds, kinds of natural products, have unique pharmacological activity, widely distributed in nature. For their novel chemical structure, a plurality of chiral center and rich function groups, they became the focus of the chemists. In this paper, we focus on the total synthesis of the threo-(±)-3,4-divanillyltetrahydrofuran and derivatives. This paper includes the following three parts:Part one is the summary, we introduced the current study status and research progress of lianan compounds about its classification, biological activities and physicochemical properties. This section expound about the research on synthesis of tetrahydrofuran lignans related in this thesis. Then we put forward the research topic combined with the current research situation.The second part introduced the total synthesis of threo-(±)-3,4-divanillyltetrahydrofuran and its derivatives. In this thesis, the target product was obtained, starting from vanillin and diethyl succinate, via Stobbe condensation, esterification, reduction and condensation reactions. By this synthesis method we also get dilignan(±)-1-O-feruloyisecoisolariciresinol.Part three, considering the mechanisms of each steps and important factors, a simple and efficient process was achieved after doing a lot of contrast experiments, in which reacting conditions,including temperature, solvent, weight ratio, reaction time,and operating methods was studied to improve the yield and the quality of products and to determine the optimal reacting condition.
