| Objective:In this paper, we reveal the role of the Buxin prescription in promoting hematopoietic function by research the influence of Zhigancao Decoction on the activity of the factor associated with hematopoiesis of the syndrome model mice, thereby more comprehensive comprehending the basic theory of TCM-’Heart is responsible for production of blood’, and it also provides more research methods and treatment ideas about aplastic anemia, tumor, etc.Method:150mice were randomly divided into5groups. According to the daily75g/kg restricted diet, except the normal group, other4groups bloodletting0.2ml and swimming20min every other day. Gavage started at the seventh day, the drug groups were given the corresponding reagents, and the normal group and the control group were given the equal amount of saline. At the same time, Continue to make mole until the experiment ends. At the third day, the seventh day and the fourteenth day after gavage, selecting10mice randomly and analyzing the peripheral hemogram of the blood sampled by eyeball extirpating, then broking neck and obtaining one side of the femur to make the pathology section. We used immunohistochemistry method to test the change of CD34, Factor Ⅷ-related antigen, EPO, SCF.Results:1. The role of Zhigancao Decoction in promoting peripheral hemogram:Compared with the normal group, the red blood cells and hemoglobin in the peripheral blood of mice reduced significantly (P<0.01) after making mole. There is no difference between the drug group and the control group after3days’gavage, and it is no statistically significant. Compared with the control group, the RBC and Hb of the drug group rose and are obvious different (P<0.01) after7days’gavage. There is no difference in the drug group, the control group and the normal group after14days’ gavage.After3days’gavage, the hemoglobin in the peripheral blood of other4making mole groups is obvious decreasing and significant different compared with the normal mice. After successful making mole,3days’gavage cannot promote the rise of Hb. After7days’gavage, the Hb of3gavage groups has increased, especially the Zhigancao Decoction group and Yougui Decoction group has obvious increased, and compared with normal group, the rise of heji group has no statistically significant. After14days’gavage, the difference between the mole group and the normal group is obvious, the Hb of3gavage groups is higher than the mole group.Compared with the normal group, the leukocyte of the mice’s peripheral blood trended to increasing, but it is no statistically significant. After3,7, and14days’gavage, the WBC of the mice of5groups is no statistically significant. After3and14days’gavage, the PLT in the peripheral blood of the mice of5groups is no statistically significant. The PLT in the peripheral blood mice with7days’gavage is some increasing, the difference of other4groups is no statistically significant.2The influence of Zhigancao Decoction’s on EPO, SCF in peripheral blood:By using ELISA method to test EPO in peripheral blood, we found that it is no obvious different between the gavage group and normal group at the third day and seventh day after gavage., and it is no statistically significant.By using ELISA method to test SCF, we found that the number of SCF in peripheral blood of the drug mice after3and7days’gavage increased greatly compared with the control group, it is statistically significant.3The influence of Zhigancao Decoction’s on the expression of CD34, factor Ⅷ-related antigen, SCF, EPO in bone marrow:The test after3days’gavage, we found the CD34in bone marrow of the4mole groups decreased compared with the normal group, the difference is obvious. And the test after7days’gavage, we found the CD34in bone marrow of the4mole groups is less than the normal group. And the test after14days’gavage, the difference among the3gavage groups is no statistically significant.The test of EPO in bone marrow, after3days’gavage, Yougui Decoction group is higher than the normal group, and the difference between the other4groups is no statistically significant. After7days’gavage, the difference between the5groups is no statistically significant. After14days’gavage, the difference between the3gavage groups is no statistically significant.The test of factor Ⅷ-related antigen in bone marrow, after3days’gavage, the mole group and the normal group have obvious different, the factor Ⅷ-related antigen in the Zhigancao Decoction group is higher than the mole group. After7days’gavage, compared with the mole group, the3gavage groups all increased, the Zhigancao Decoction group and Yougui Decoction group increased obviously. After14days’gavage, the3gavage groups all increased, especially the Zhigancao Decoction group and Yougui Decoction group increased obviously.The test of SCF in bone marrow, after3days’gavage and7days’gavage, the number between the5groups is no statistically significant. After14days’gavage, the heji group is higher than the mole group, the difference between the other4groups is no statistically significant.Conclusion:1By analyzing the peripheral hemogram of mice, we consider Zhigancao Decoction can promote the multiplication of the red blood cells and hemoglobin in the peripheral blood under certain conditions.2By using ELISA method to test peripheral blood, we found that the number of SCF in the peripheral blood of the mice with3different dose groups increased. We think Zhigancao Decoction and Yougui Decoction can improve the peripheral hemogram of the blood deficiency mole mice, and improve the secrete of SCF to activate dormant stem cells and promote its growth. At the same time, they can adjust the micro environment of the body, to offer the better environment to stem cells, and stimulate the bone marrow to hematopoietic.3We found the secrete of EPO cannot be improved by the medicine used in the experiment in both the peripheral blood and the bone marrow. Zhigancao Decoction and Yougui Decoction can improve the secrete of the factor Ⅷ-related antigen of the blood deficiency mole mice. |
