Dioscoreae Rhizoma is the Dioscorea opposita Thunb.dry rhizome."Chinese Pharmacopoeia" records it can nourish the stomach, invigorate spleen and generate saliva profit lung. It can be used for the treatment of splenasthenic, chronic diarrhea, kidney spermatorrhea, deficiency-heat and so on. Study found that it contained large amounts of myxoid stroma. Myxoid stroma can increase the number of leukocytes in the blood, and strengthen the capacity of leukocyte phagocytic. Thus, it can be used as the tumor of chemotherapy adjuvants. Dioscoreae Rhizoma can be eaten alone or with other food materials. Therefore, the deep development of Dioscoreae Rhizoma has practical value.The main active ingredient is the Dioscoreae Rhizoma polysaccharide. It can be widely used in the fields of medical and health care. It has the function of treatment of diabetes, anti-tumor effect, anti-aging and increasing the resistance of body and so on. This study further explores extraction of polysaccharides from Dioscoreae Rhizoma. It used water extraction and alcohol precipitation method for extracting of the Dioscoreae Rhizoma polysaccharide and under the basis of factors including extraction temperature, extraction time and additive water amount to design orthogonal.Eventually,the yield was the orthogonal screening indicators.Experimental results showed that the optimum process was that the extraction temperature was90℃, the extraction time was3hours and the solid-liquid ratio was1g:25mL. The results are used for researching Dioscoreae Rhizoma polys accharides and controling quality.The collected six kinds of commercially available of Dioscoreae Rhizoma, and did systematical researeh to quality control factors of them. Firstly, the carried on micro and TLC identification of their powders. Secondly, the determined of which water, total ash, acid insoluble ash, extract, polysaccharide, allantoin, trace elements and heavy metals concentrations.(1)Use the ultraviolet-visible spectrophotometry to determine thecontents of Pachymaran in the Dioscoreae Rhizoma. The rusults showted that the contents between1.82%and2.40%.(2)Set up a flame atomic absorption spectrometry to determine the contents of Mg, Fe, Zn and Mn. The results showed that the order was Mg>Fe>Zn>Mn. The recovey was between95.50%and103.5%and RSD between0.67%and4.35%.(3)Set up the wet digestion-atomic absorption spectrometry to determine the contents of heavy metals Cu, Pb. Cd and Cr in the samples. The results showed that the contents of Cu, Pb and Cd were not exceeded and the experimental results corresponded analysis requirements.(4)Use HPLC method to determine the contents of allantoin in Dioscoreae Rhizoma. The results showed that the contents of the samples were between0.1125%and0.2840%. The results of this study can provide scientific reference for the quality control and reasonable utilization of Dioscoreae Rhizoma.
