| Backgroud and Objective:In recent years, the adverse reaction of traditional Chinese medicine at home and abroad caused by the reports, so that more and more people doubt the safety of traditional Chinese medicine. As a medicine, it is more or less toxic. Chinese medicine even natural animals and plants as well as sources of minerals is no exception. But not because of the adverse reaction of traditional Chinese medicine to deny the clinical value of traditional Chinese medicine, and should seek for a method with the aid of scientific means to reduce adverse reactions. Rhizoma Dioscoreae Bulbiferae is the dried tuber of Dioscorea bulbifera L. As reported on the adverse reactions in recent years, greatly limited its unique curative effect in clinic. The experimental study based on the theory of traditional Chinese medicine"sweet can slow" and "detoxicity by compatibility". Combined with modern technology to study gene expression in order to achieve the purpose of easing bitter and cold of Rhizoma Dioscoreae Bulbiferae, and reducing its side-effects by the compatibility of Radix Glycyrrhizae. As the same time for safe clinical application and provide experimental basis.Methods:The first section of the paper by retrieving and analysing the clinical reports about adverse reaction of Rhizoma Dioscoreae Bulbiferae and related preparations, through the modern medical journals database. And according to the filtered into and exclusion criteria, the last of 99 clinical adverse reactions report is analyzed. I will be discussed from four aspects that are the general situation, lead to adverse reactions happen by Rhizoma Dioscoreae Bulbiferae and its different type, the main causes of adverse reactions occur and the classification and clinical manifestation.The second part of the thesis mainly by experiment means to explore if Radix liquiritiae reduce liver damage of Rhizoma Dioscoreae Bulbiferae through regulating Bcl-2/Bax, and provide the scientific basis for the clinical reasonable application of Rhizoma Dioscoreae Bulbiferae. The main contents are the detection of weight, liver index, serum biochemical indexes and through the application of reverse transcriptase chain polymerization technology, protein imprinting technique in mRNA and protein expression in mouse liver Bax detection, the amount of Bcl-2 gene in these grops that the group of Rhizoma Dioscoreae Bulbiferae, the group of Rhizoma Dioscoreae Bulbiferae combined with Radix liquiritiae with the scale of 1 than 2, the group of Radix liquiritiae and blank control group.Result:(1) The characteristics of the adverse reactions caused by Rhizoma Dioscoreae Bulbiferae and its preparation is mostly occur in women. The adverse reactions caused by airpotato yam and its preparation in most mainly in Rhizoma Dioscoreae Bulbiferae Decoction (74.7%). And the leading cause of clinical adverse reaction is too large doses of medication, followed by the duration of treatment and the patient privately medication.In addion, adverse reactions of Rhizoma Dioscoreae Bulbiferae and related preparations mainly based on liver damage is given priority to, embodied in the sclera, skin is orange, liver area pain and abnormal hepatic function indexes, etc.(2) The group of Rhizoma Dioscoreae Bulbiferae compared with blank control group, the growth of body weight decreased with statistical significance (P<0.05), and combined with Radix liquiritiae, weight gain increased relatively;(3) the group of Rhizoma Dioscoreae Bulbiferae compared with blank control group, The liver index increased significantly (P<0.01), and after combined with Radix liquiritiae and compared with blank control group, the liver indexhad significantly decreased (P<0.01);(4) the group of Rhizoma Dioscoreae Bulbiferae compared with blank control group, three biochemical indexes (ALTã€ASTã€AKP) of the group of Rhizoma Dioscoreae Bulbiferae Increased significantly (P<0.05), and after combined with Radix liquiritiae and compared with blank control group, the above three indicators were significantly reduced (P<0.05);(5) at mRNA and prote in level, the group of Rhizoma Dioscoreae Bulbiferae compared with blank control group, the expression of Bcl-2 and Bax gene were significant differences (P<0.05). The trend for mice Bcl-2 gene expression is down and Bax expression is up of the group of Rhizoma Dioscoreae Bulbiferae. And after combined with Radix liquiritiae and compared with blank control group, the expression of Bcl-2, Bax gene also were significant differences (P<0.05). After combined with Radix liquiritiae and compared with blank control group, the trend for mice Bcl-2 gene expression is up and Bax expression is down.In summary, suggested that Radix liquiritiae may by regulating the expression of Bcl-2 and Bax gene to alleviate liver damage in mice. Also verified the understanding of ancient physicians has a role in detoxification and protect the liver of Radix liquiritiae. |
