| ObjectiveTo detect the expressions of Survivin, DcR3in protein and geneexpression level respectively in human gastric carcinoma(GC) tissues. Thenexplore their effects during the occurrence and development of human gastriccarcinoma.MethodsTo select50patients with gastric cancer and50cases of correspondingadjacent cancer normal tissues, using by immunohistochemical and RT-PCRmethod to detect organizations of Survivin and DcR3respectively in the levels ofprotein and gene expression, and carrying out the relevant research.Results(1)The protein expression of Survivin in gastric cancer is mainly localized inthe cytoplasm and nucleus, The positive expression rate is68.0%, theexpression of adjacent normal tissue rate is2.0%,the difference has remarkablestatistics significance (P<0.01). The protein expression of DcR3in gastriccancer is mainly localized in cytoplasm and membrane, The positive expressionrate is72.0%, the expression of adjacent normal tissue rate is6.0%,thedifference has remarkable statistics significance (P<0.01).(2)The expression of Survivin mRNA levels(0.745±0.058) in the gastriccancer group were much higher than in normal gastric mucosa groups(0.281±0.125),the difference was statistically significant(P<0.05). The expression of DcR3mRNA levels(0.658±0.072) in the gastric cancer group were alsoSignificantly higher than normal gastric mucosa groups(0.153±0.069), and thedifference was statistically significant(P<0.05).(3)In two levels of both protein and mRNA, The expression level of Survivinand DcR3are related to the gastric cancer, depth of invasion, tissuedifferentiation, lymph node metastasis and TNM stages (P<0.05). Regardless ofage, gender and size of tumor. Analyzed by Spearman relevant inspectionshows that the expression of two-factor is positively correlated(P<0.01).Conclusions(1)The high status expression of Survivin and DcR3factor in gastric cancertissues is positively correlated, and closely related to gastric tissue differentiation,lymph node metastasis and TNM stage, promoting there may be somecoordination role in gastric carcinogenesis, development, invasion andmetastasis process.(2)Survivin and DcR3both can be used as indicators of biological behavior ofgastric cancer and clinical prognosis, The accuracy is even higher with combinedetection. |
