| Pigment epithelium derived factor (Pigment epithelium-derived factor, PEDF) is a glycoprotein belonging to the serine protease inhibitor superfamily. Was originally purified in a human retinal pigment epithelial and with potential neuronal differentiation[1]. It has been confirmed that, PEDF in the cell culture and animal models can inhibit angiogenesis and can inhibit retinal endothelial cell growth, migration, and ischemia-induced angiogenesis. PEDF are expressed in a variety of tissues, including vascular endothelial cells, inflammatory cells and fat cells [2]. The study found that the incidence of metabolic diseases PEDF in the serum increased with the crowd is proportional unclear, but in the serum expression levels of PEDF and atherosclerosis [3]. This project intends to study the PEDF whether as atherosclerotic disease independent detection indicators for further clinical diagnosis and treatment.1PurposeThe relationship between research carotid atherosclerotic plaque stability and serum PEDF level of cerebral infarction observed PEDF instability of atherosclerotic plaques and cerebral infarction, a potential serum markers.2Methods2.1carotid atherosclerotic plaques in the nature the Philips ATL-HDI5000color Doppler ultrasonography (ultrasound probe frequency of5~12MHz), the routine examination of bilateral common carotid artery (CCA), carotid bifurcation (BIF), internal carotid artery (ICA), vertebral artery (VA). Carotid artery plaque diagnostic criteria for carotid intima thickness (IMT)≥1.2mm, local uplift highlight to the tube [6]. Ultrasound features fractal type, namely (1) the unstable plaque:Local hypoechoic, isoechoic or mixed echo the soft spots;(2) stable plaque:high, strong echoes with or without acoustic shadowing morphea. Cerebral infarction group were divided into unstable plaque group and stable plaque group and the group without plaque three subgroups.2.2serum PEDF levels of detection All patients had fasting blood admission extraction2d6ml, centrifuged to obtain serum, placed at-70℃kept under test, serum PEDF solid phase double antibody sandwich enzyme-linked immunosorbent assay (ELISA) was measured The kit was purchased from Beijing the Bole life science development Co., Ltd..3ResultsThe3.1CI group and the control group plaque detection rate of cerebral infarction group were detected in the the plaque40cases,46cases, including18cases of unstable plaque plaque detection rate of86.96%, accounting for39.13%. CCCI group were detected in25cases,15cases of plaque detection rate of60%; including three cases of unstable plaque, accounting for12%.3was detected in the plaque, plaque detection rate of20%in the control group of15patients, unstable plaque. Plaque composition than three groups were significantly different, a higher incidence of carotid artery atherosclerotic plaques in the cerebral infarction group and unstable plaque.3.2serum the PEDF level of cerebral infarction serum PEDF levels were significantly higher than the CCCI group and the control group. The subgroup of patients serum PEDF level there are significant differences, the unstable plaque infarction group was significantly higher than the stable plaque infarction group, stable plaque infarction group was significantly higher than the CI group without plaque. In addition, no plaque cerebral infarction group PEDF levels were also significantly higher than the the CCCI group and the control group.Conclusion:Serum PEDF expression levels can be used as indicators of the severity of the patients with a monitoring for further clinical application. |
