Objective: This experiment discusses the expression of HSP90α, HSP90β protein andmRNA in human gastric carcinoma tissues on the cellular and molecular level andanalyzes their relationship with the gastric carcinoma clinical pathological features inorder to explore their relationship with the biological behavior of gastric carcinoma.Methods:(1) Using immunohistochemical S-P method detects gastric carcinomatissues and the expression of HSP90α and HSP90β protein (HSP90α:70cases,HSP90β:75cases) in the adjacent normal tissues (30cases).(2)Reversingtranscription-polymerase chain reaction (RT-PCR) method was applied to detect32cases of gastric carcinoma tissues and the expression of HSP90αmRNA andHSP90βmRNA in the adjacent normal tissues.(3) Both results ofimmunohistochemistry and RT-PCR were combined to clinical and pathological datato analyze the relationship between HSP90α, HSP90β and the occurring, development,invasion and metastasis of gastric carcinoma. Results:(1) In gastric carcinoma andadjacent normal tissues, the positive expression rate of HSP90α protein were82.9%(58/70) and43.3%(12/30) respectively, so there was significant statisticalsignificance between the differences of two groups (P <0.05). There was alsostatistical significance that the positive expressions of HSP90α protein werecorrelated with the tumor depth of invasion, the lymphatic metastasis and TNM stage(P <0.05), but there was no specific association with the gender, age, and pathologicalgrades of the patients. The expressions of HSP90αmRNA in gastric carcinoma wassignificantly higher than the adjacent normal tissues (P <0.01), which is related to thegastric cancer differentiation, the lymphatic metastasis and TNM (P <0.05),but notrelated to the depth of tumor(P <0.05).(2)In gastric carcinoma and adjacent normaltissues, the positive expression rate of HSP90β protein were80.0%(60/75)and30.0%(13/30)respectively, so there was significant statistical significance between the differences of two groups (P <0.05). The positive expressions of HSP90β proteinwere correlated with the differentiated degree of tumor, the lymphatic metastasis andTNM stage (P <0.05), so their differences have statistical significance. There was nospecific association with the gender, age, depth of invasion and the tumor size of thepatients. The expressions of HSP90βmRNA in gastric carcinoma was significantlyhigher than the adjacent normal tissues (P <0.01), which is related to thedifferentiated degree, the TNM stage and lymphatic metastasis (P <0.05), but notrelated to the depth of invasion (P <0.05). Conclusion:(1) The expressions ofHSP90α and HSP90β protein in the gastric carcinoma were closely related to theinvasion, metastasis, clinical stages and the differentiation of tumor, and HSP90α andHSP90β served as useful indicators can determine the malignant of gastric carcinoma.(2) Both the high expressions of HSP90αmRNA and HSP90βmRNA, which areclosely related to the occurring and development of gastric carcinoma, may haverelationship with the transcriptional regulation in the gastric carcinoma tissues. |
