| Purpose To study the expression of ZEB-1 and c-jun and its influence of occurrence and development in gastric carcinoma.The model of silencing expression of ZEB-1 gene was established in human gastric carcinoma cells BGC-823 and to study its effects and mechanism on gastric carcinoma cell proliferation,migration,invasion.Methods The expression ZEB-1 and c-jun were assessed in 100 specimens of gastric carcinoma and their tissue adjacent to cancer by immunohistochemistry(IHC).We build interference sequence of si RNA to silence ZEB-1 gene,which use it to transfect gastric carcinomacells.The gene silencing efficiency of ZEB-1 was measured by reverse transcription PCR.Cell proliferation was examined by CCK-8 assay; Migration and invasive capability were measured by transwell chamber.Results The positive rates for ZEB-1 and c-jun and E-cadherin expression in gastric carcinoma were 81%,70%,35%and in tissue adjacent to cancer were17%,25%,100%. The expression of ZEB-1 and c-jun in gastric carcinoma were higher than those in their tissue adjacent to cancer(P<0.05).ZEB-1 expression was closely correlated with the degree of tumor differentiation,depth of invasion,lymph node metastasis and TNM stage( P <0.05),which were not correlated with patient ′ s age,sex and tumor length.c-jun expression was closely correlated with the degree of tumor differentiation(P<0.05), which were not correlated with other pathological features.The expression of ZEB-1 was negatively correlated with the expression of E-cadherin and was positively correlated with the expression of c-jun in gastric carcinoma(P<0.05).Patients with high expression of ZEB-1 and c-jun had lower five-year survival rate than patients with negative expression.After transfected BGC-823 cells with si RNA, the expression of ZEB-1-si RNA3 in the m RNA levels was significantly inhibited by PCR.CCK-8 assay detected that the ability to proliferation of gastric carcinoma cells was not significantly different(P>0.05).The results of migration and invasion by transwell chamber showed that the capacity of experimental group was lower than that of the negative control group and blank control group(P<0.05).Conclusion ZEB-1 and c-jun were closely correlated with occurrence and development of gastric carcinoma.They can be a index of judging prognosis of gastric carcinoma. ZEB-1 gene play an important role in promoting gastric carcinoma invasion and metastasis and can become a new target to control invasion and metastasis of gastric carcinoma. |
