Effect Of Nourishing Kidney And Activating Blood Recipe Of Traditional Chinese Medicine On Podocyte Of Adriamycin Nephropathy Mice

Objective To investigate the effect of nourishing kidney and activating blood recipe of traditional Chinese medicine on the expression of nephrin in renal tissue of mice with adriamycin induced nephropathy.Methods The nephropathy model of focal segmental glomerulosclerosis(FSGS) was induced by injection of adriamycin into the tail vein. After1week, twenty four male rats were randomly divided into normal control group, model group and treatment group. The levels of body weight, Albumin (ALB) and urinary albumin creatinine ratio (UACR) were measured. Furthermore, the morphological changes of renal tissue were observed under microscope. The protein expression of desim in kidey tissures were determine by immunohistochemical.The mRNA of nephrin and desmin in renal tissues were determined by RT-PCR.The protein expression of nephrin in kidey tissures were determine by immunofluorescence labeling.Results The levels of body weight and ALB were significantly increased, the level of UACR was significantly decreased in normal control group and treatment group when comparing with model group (P<0.01). There was intense positive expression of nephrin and lower protein expression of desmin in glomerular mesangial region in normal control group and treatment group. There was significantly higher expression of nephrin mRNA, lower expression of desmin mRNA in treatment group than in model group (P<0.01).Conclusion The method of nourishing kidney and activating blood recipe of traditional Chinese medicine reduces the proteinuria of the FSGS nephropathy models, which might be closely related to the decreased expression of nephrin and the increased expression of desmin.
