Objective:Through experimental study,the effect of leech on blood,urine and renal tissue collagen and TGF-beta 1 was observed in rats with segmental glomerulosclerosis(FSGS),and the relevant theoretical basis was provided for the treatment of FSGS by leech.Methods: The model of adriamycin nephropathy was established by unilateral nephrectomy and intravenous injection of adriamycin twice.It was divided into four groups:treatment group,model group,sham operation group and blank group.Changes of general status,serum total cholesterol,plasma albumin,urinary microalbumin/urinary creatinine in rats were observed dynamically.HE staining was performed on kidney sections to observe the pathological changes of renal tissue in rat model group of adriamycin nephropathy.Ultrastructure of kidney was observed by electron microscopy.Immunohistochemical two footwork,real-time fluorescence quantitative method to observe Ⅳ collagen type and the change of the expression of TGF-beta 1.Results:1.Leech treatment group can improve the general state of adriamycin nephropathy rats;Serum total cholesterol,urinary microalbumin/urinary creatinine were lower in the treatment group than in the model group,and plasma albumin was higher in the model group than in the model group,with statistically significant difference(P<0.05).2.Leech treatment group can reduce the pathological damage of FSGS;3.The immunohistochemical results show that compared with leech treatment group and the blank group,model group Ⅳ collagen type in glomerular endothelial cells and mesangial cells and epithelial cells express,the difference was statistically significant(P < 0.05),compared with model group,leech treatment group,the expression of collagen type Ⅳ decrease,the difference was statistically significant(P < 0.05);Compared with leech treatment group and the blank group,model group TGF-beta 1 on glomerular mesangial cells express,the difference was statistically significant(P < 0.05),leech treatment group,the expression of TGF-beta 1 compared with model group,expression is abate,the difference was statistically significant(P < 0.05).4.Rt-pcr results show that compared with leech treatment group and the blank group,model group,the expression of increasing,the difference was statistically significant(P < 0.05),compared with model group,collagen type Ⅳ expression of leech treatment group decreased,the difference was statistically significant(P < 0.05);Compared with leech treatment group and the blank group,model group TGF-beta 1 mrna expression increases,the difference had statistical significance(P < 0.05),compared with model group,leech treatment group TGF-beta 1 mrna expression decrease,the difference was statistically significant(P < 0.05).Conclusions:1.Leech can reduce the excretion of microalbumin in the urine of FSGS rats,reduce the total cholesterol content,and improve the plasma albumin.2.Leech can reduce renal pathological damage;3.Leech is probably by reducing the expression of TGF-beta 1 and collagen Ⅳ,thereby reducing the accumulation of ECM,delay the progress of FSGS.
