Objective:To investigate and observe the effect of Korean medical prescription Xiangshalizhong decoction on the peptic ulcer healing process of gastric juice quantity, pepsin activity, the content of serum nitric oxide (NO), serum gastrin (GAS) and PH value of gastric juice, and primarily explore the possible mechanism.Methods:(1) Sixty rats were randomized into6groups, including Normal Control group, Gastric Ulcer Model group, Positive Control group (Ranitidine), Xiangshalizhong decoction high middle and low dose group.(2) Acetic acid-induced gastric ulcer rat model was taken into use, two days after gavage14days, High-dose group gavage12. Og/kg, Middle-dose group gavage6. Og/kg, Low-dose group gavage3. Og/kg, Normal Control group and Gastric Ulcer Model group gavage0.9%sodium chloride injection2ml, Positive Control group gavage Ranitidine suspension30mg/kg.(3) Gavage14days of treatment, to fast and cannot refrain from water for24h, intraperitoneal anesthesia, obtain blood from heart and collect gastric juice, then use high-speed centrifuge separation and preserved at-20℃.(4) Observe the gastric mucosa in general view, detecte gastric juice quantity, pepsin activity, the content of serum nitric oxide (NO) and serum gastrin (GAS), PH value of gastric juice.(5) Use SPSS13.0software package for processing statistical analysis of experimental data.Result:(1) Xiangshalizhong decoction high middle and low dose groups can reduce the ulcer index (P<0.05), there is significant difference;(2) In comparison of xiangshalizhong decoction high and middle dose groups with model group, gastric juice volume was obviously reduced (P<0.01), manifesting obvious difference;Xiangshalizhong decoction low dose group compared with model group (P>0.05), showed no statistical significance;(3)Xiangshalizhong decoction high and middle dose groups compared with Gastric Ulcer Model group,pepsin activity was significantly reduced, bearing distinct difference (P<0.01);Xiangshalizhong decoction Low dose group and Ranitidine group compared with Gastric Ulcer Model group, pepsin activity was also significantly reduced(P<0.05), which displays conspicuous difference. In comparison of the Xiangshalizhong decoction high and middle dose groups with Ranitidine group, significant reduction is seen in pepsin activity (P<0.01).(4) Xiangshalizhong decoction High dose group and Ranitidine group compared with Gastric Ulcer Model group, the content of serum NO was obviously increased, bearing obvious difference (P<0.05); In comparison of Xiangshalizhong decoction Middle dose group with Gastric Ulcer Model group, the content of serum NO was also obviously increased (P<0.01), displaying conspicuous difference.(5)The volume of GAS in the Xiangshalizhong decoction Middle dose group and Ranitidine group were significantly lower than Gastric Ulcer Model group (P<0.01), and the high and low dose groups were lower than Model group obviously (P<0.05), which displaying apparent difference;(6)In comparison of Xiangshalizhong decoction Low dose group and Ranitidine group with Model group, PH value of gastric juice was decreased obviously (P<0.05), bearing marked difference; While the high and middle dose groups compared with Model group, showed no statistical significance (P>0.05)Conclusion:Korean medical prescription Xiangshalizhong decoction can promote the healing of acetic acid-induced gastric ulcer rat significantly. The mechanism may be related to reduce gastric secretion, pepsin activity and serum gastrin, to increase nitric oxide volume in serum. |
