Part1Association of MAOA gene and LIN28B gene polymorphismwith digit ratio of Ningxia populationObjective:this study was to investigate the correlation between two different genespolymorphisms and digit ratio of Ningxia region population. The genespolymorphisms including the rs6323and the promoter region of30bp nucleotidepolymorphisms (30-VNTR) of monoamine oxidase (MAOA) gene and rs314277ã€rs314276of LIN28B gene, to verify whether this four polymorphisms are thebiological basis of human finger digit ratio development. And to to investigate thecorrelation between the two sites of LIN28B gene and the data of the human bodyconstitution of Ningxia region population.Methods:All671Subjects were from Ningxia Medical University, including294cases of boys, of which136cases of the Hui and158cases of the Han;377cases ofgirls, including the182cases of Hui and the195cases of Han. the average agewas19.85±1.4years. Digit Ratio was according to the Manning criteria (Manning JT,"Digit Ratio" Rutgers University Press,2002), using a digital camera extract subjects’left and right hands photographs, remove the finger injuries samples, record the ageand other relevant parameters, the sample datas were stored in the computer, usingPhotoshop software processing flag anatomical points of each finger, print them anduse electronic vernier calipers to measure, the measurement precision was0.01mm.And recorded the length of four fingers of each hand, all subjects were measuredtwice. Calculate the ratio of the left and right finger length ratio (2D:3D,2D:4D,2D: 5D,3D:4D,3D:5D,4D:5D). The heightã€Weightã€waist circumference and HipCircumference were measured by using the method of Shao Xiangqing reported. Thers6323site polymorphism were examined by the Allele-Specific polymerase chainreaction(AS-PCR) methods; The30-VNTR site polymorphism were examined by theelectrophoresis after polymerase chain reaction methods. The SNPs of LIN28B genewere examined by the Gene Sequencing methods. The genotype distribution of thefour SNPs’ Hardy-Weinberg equilibrium were tested by using The SHEsis software.using the SPSS18.0statistical software to establish the database and to process andanalysis all the datas. The comparison of genotype and allele frequencies werecalculated by direct counting, body mass index (BMI), waist-hip ratio (WHR) anddigit ratio measurement with Lin28b gene’s SNPs in gender, ethnic was usedindependent sample t-test. The MAOA gene SNPs were examined by usingindependent sample t-test in boys,and by using One-way analysis of variance(One-way ANOVA) in girls.Results: Our study has found that in men and women, the2D:4D was higher in lefthand than in the right hand (P <0.001). The height, weight, waist circumferencedistribution in different gender was statistically significant in Ningxia people(P <0.01),the weight distribution was statistically significant in different ethnic of Ningxiapeople(P <0.01). The waist-to-hip ratio distribution difference was statisticallysignificant in different ethnic of Ningxia people(p <005),but the distribution of bodymass index was not statistically significant (p>005). We found that there was nosignificant difference between the two SNPs in Lin28b gene with the left and righthand2D:4D, height, weight, waist circumference, hip circumference, waist-to-hipratio and body mass index of Ningxia people. There was no significant differencebetween the two SNPs in MAOA gene and the left and right hand2D:4D (p>005).Conclusion: there was no significant difference between digit ratio2D:4D and MAOA gene and LLIN28B gene polymorphisms in Ningxia people. In our presentstudy, we found that the left hand2D:4D ratio was longer than that of the right handin both males and females.We have found that the left hand2D:4D ratio was shorterin male than in female, we did not found the right hand2D:4D ratio was shorter orlonger in male than in female. The waist-hip ratio distribution was significantdifference in Ningxia different ethnic population.The BMI was no significantdifference between the different ethnic and gender of Ningxia populations. We did notfound there was significant difference between Lin28b gene polymorphisms and digitratio, height, weight, waist circumference, hip circumference, waist-to-hip ratio inNingxia population. Part2Study on the MAOA gene polymorphisms in Ningxia Huiand Han ethnic peopleObiective:To study the frequencies of allele and genotype distribution of the MAOAgene polymorphisms in the Hui and Han people in Ningxia, and to analyze thedistributions of MAOA gene polymorphisms among different places and race.Methods: The rs6323site polymorphism were examined by the Allele-Specificpolymerase chain reaction(AS-PCR) methods; The30-VNTR site polymorphism wereexamined by the electrophoresis after polymerase chain reaction methods. They bothin309males(143Hui and166Han) and414females(187Hui and227Han).Results:The genotypes frequencies and allele gene frequency of MAOA gene inNingXia Hui ethnic people was: ale30-VNTR (H:43.36%; L:56.64%); rs6323(T:44.06%; G:55.94%); Female30-VNTR HH:16.0%; L:66.8%; L:17.1%;:49.5%;L:50.5%); s6323TT:17.6%; G:46.5%; G:35.8%;:40.9%;:59.1%); The genotypesfrequencies and allele gene frequency of MAOA gene in NingXia Han ethnic peoplewas: Male30-VNTR (H:49.4%; L:50.6%); rs6323(T:49.4%; G:50.6%);emale30-VNTR HH:19.4%; L:41.0%; L:39.6%;:39.9%;L:60.1%); s6323(TT:17.2%;TG:44.9%; GG:37.9%;T:39.6%; G:60.4%).Conclusion:30-VNTR site’s genes distribution is significantly different in theHui and Han ethnic of NingXia female population. The differences of d istribution traits of genotype and allele frequencies in MAOA gene indicate that this two SNPshave genetics characteristic in different ethnics. |
