Nowadays artificial abortion and medical abortion were generally applied to termination of pregnancy, and the complications, for instance, Uterine perforation, intrauterine adhesion, and decidua of fundus uteri were generally occurred. Patients have to face procedural safety and unknown pain level in an anxiety or horror mode. And repeatedly abortions would lead to serious problem of female fertility. In past several years, Gel acupoint injection was a new method of traditional acupoint injection. It is a combination of acupoint injection and acupoint catgut embedding. It’s also used as an development of acupuncture therapy. Gel-acupoint injection might be a newly method of termination of pregnancy due to its degradation, convenience, delayed release, acupoint stimulation and various drug bearing.Objective: This study established mouse models of earlier pregnant period. Though observing the changes of Estradiol(E2), Luteinizing Hormone(LH) and the expression of ER-αmRNA were used to assess the role of Gel-acupoint injection in pregnant fertility.It’s expected to explore the possible mechanism of Gel-acupoint injection in the treatment of termination and provide evidence and data reference for the Gel-acupoint injection.Method: Six weeks old healthy adult Kunming female mouse(totally50mouse) were divided into five groups randomly: normal control group, model group; acupuncture group, gel-acupoint injection groups and mifepristone group.Each group was included ten mouse. The pregnant mouse models were made by mating at the ratio of2:1. After modeling, acupuncture, gel-acupoint injection, mifepristone and were applied to treat in acupuncture group, gel-acupoint injection group and mifepristone group. The changes of Estradiol(E2), Luteinizing Hormone(LH) were tested by the test of ELISA. And the changes of the expression of ERαmRNA was tested by the test of Real-PCR.Result:1.In comparison with normal control group after modeling, the Estradiol(E2) increased significantly (P<0.01) in the model group. In comparison with normal control group after modeling, the Estradiol(E2) decreased in acupuncture group, gel-acupoint injection group and mifepristone group.In comparison with model group, there was no significant difference showed in acupuncture group (P>0.05), but there was a significant difference in gel-acupoinl injection group (P<0.05).In comparison with model group, the Estradiol(E2) decreased significantly in mifepristone group (P<0.01), In comparison with acupuncture group, the Estradiol(E2) decreased significantly in acupuncture group (P<0.05)2.In comparison with normal control group after modeling, the luteinizing hormone(LH) increased significantly (P<0.05) in the model group.In comparison with normal control group, the luteinizing hormone(LH) increased in the acupuncture group, but there was no significant difference was shown (P>0.05).In comparison with normal control group, the luteinizing hormone(LH) increased significantly in the gel-acupoint injection group (P<0.05).In comparison with mifepristone group, the luteinizing hormone(LH) decreased significantly in both acupuncture group and gel-acupoint injection group (P<0.05). In comparison with model group the luteinizing hormone(LH) decreased significantly in mifepristone group (P<0.01)3. The result of Real-time PCR tests showed that there was no significant difference of expression of ER-mRNA between all groups (P>0.05)。In comparison with model group, the expression of ER-mRNA decreased, but no significant difference (P>0.05).Conclusion:Gels implanted Shimen (RN5) by reducing the implantation mouse serum E2and LH content, change the expression of ER, the impact of the the HPO axis of the regulatory mechanisms of the pregnant rats, undermine the the implantation desired balance within the environment, to achieve the anti-the effect of early pregnancy. |
