| Lysimachia christinae Hance is a kind of commonly used traditional Chinese medicines, which eliminates dampness and heat, Tonglin, and reduces the swelling. The2010edition of "Chinese Pharmacopoeia" defined the whole dried grass of Lysimachia christinae Hance as the medical material.To comply with the "standard material calibration of standards and establish the technical specifications", which is one topic of the "Eleventh Five-Year National Science and Technology Support Program-Major Drug Discovery research project"--"research and development of standards for Chinese medicine material technology platform", This paper elaborated the study on the calibration of technical standards of Lysimachia christinae Hance reference drug, and laid a foundation of speeding up the construction of the evaluation system standards for Chinese medicine material and structuring the world-class domestic leading technology platform of Chinese medicine material.The results were as follows:1.The main origin and commodity survey results showed that Lysimachia christinae Hance was mainly produced in Jinyan of Leshan in Sichuan province. It was mostly wild and significantly-plentiful.Lysimachia christinae Hance sold by the Chinese medicinal herbs market in Chengdu lotus pond and used by Chinese medicine shops and medical units in Sichuan Province mostly come from Jinyan county. They were also sold to Chinese medicine markets all over the country. The amount in other places of commodities was not much. Most places market their own products. Some enterprises in Tongliang county also had cultivation, for personal use.2.The dynamic changes of the main component of Lysimachia christinae Hance during the different growing season were firstly studied. The results showed that in the eight batches of samples of different harvest times, the amount of the extract was higher in mid-April to mid-June. The content of quercetin and kaempferol, as well as the total amount of quercetin and kaempferol were higher in mid-April to mid-July. After August, the content of quercetin and kaempferol were significantly reduced, and the extract was also decreased. This study provided a reference to confirm the best harvest time of Lysimachia christinae Hance control. 3.The morphological and histological comparative observation and identification among the original plant, the local medicinal materials and the commercial product of Lysimachia christinae Hance in different area were taken. Combined with the literatures and the current version of "Chinese Pharmacopoeia", the original plant morphology, medicinal properties and microscopic characteristics were described. About the original plant morphology, the identification were taken mainly by the growth-way of leaves and flowersã€the glandular line on the leavesã€hair conditions, etc; About the medicinal properties, the identification were taken mainly by the color, surface coat or not, the growth-way of leaves and flowers,even the black or brown stripes on leaves; About the microscopic characteristics of the stem cross-section, we distinguished and identified mainly by the proportion of various parts and the distribution of the secretory ducts; When authenticated from the surface view and powder mirror picking leaves, the conditions of epidermal cells, stomatal and non-glandular hairs, glandular hairs, the secretion road, the pigment block were mainly used.4.The identification of flavonoid aglycones and flavonoid glycosides in Lysimachia christinae Hance was firstly taken by the Thin-Layer Chromatography. The spots of flavonoid aglycones could be detected more when using toluene-ethyl acetate-formic acid (5:4:1) as the developing solvent. The spots were clear, centred and had no tail. By comparing the flavonoid aglycones TLC spots of10batches of Lysimachia christinae Hance from different origin, five spots with the Rf values0.53,0.61,1.01,1.05and1.40were common.The latter could get good separation effect when ethyl acetate-formic acid-water (8:1:1) was used. The spots were more and clear, the Rf value was moderate and the flavonoid glycosides could separated better, we compared the thin-layer characteristics map of flavonoids glycosides in10batches of different origins of Lysimachia christinae Hance, and determined the common spots, the RF values were0.13,0.23,0.34,0.43,0.62,0.70,0.83,0.92,0.98.The similarity was calculated and evaluated by the Cosine formula. The different origins of Lysimachia christinae Hance’s samples had a high similarity. But the related plants of Lysimachia congestiflona Hemsl had the lower similarity and the TLC spots of flavonoid aglycones and flavonoid glycosides had a significant difference.5.The HPLC analysis method of Lysimachia christinae Hance was established first time. The method was stable, reliable and reproducible. It could indicate the main condition of the constituents in Lysimachia christinae Hance comprehensively and directly. By comparing the HPLC results of10batches of Lysimachia christinae Hance from different origins, eight characteristic peaks were identified as the characteristic HPLC fingerprints. Three of them were identified as rutin, hyperoside and quercitrin. The HPLC fingerprint of Lysimachia christinae Hance’s samples from different origins had the high similarity. But the related plants of Lysimachia congestiflona Hemsl had the lower similarity, indicating that the ingredient between them were different, and showed the characteristics of the HPLC fingerprint.6. Ten batches of Lysimachia christinae Hance originated in different places were examined from the impurities, moisture, total ash and acid insoluble ash according with the current edition of the "Chinese Pharmacopoeia". According to the test data, combined with the relevant provisions of the current version of "Chinese Pharmacopoeia", and considered the reference medicinal material requirements, the standard of its’limitation was that:impurities should not be more than8.0%moisture should not be more than13.0, total ash should not exceed13.0%, acid insoluble ash should not be more than5.0%.We determined the content of herbs extract, quercetin and kaempferol in10batches of different origins of Lysimachia christinae Hance counterparts medicines and commercial medicines. According to the measured data, combined with the relevant provisions of the current version of "Chinese Pharmacopoeia", and considered the reference medicinal material requirements, the standard of its’ content limitation was that:The extract could not be less than8.0%, the total amount of quercetin and kaempferol could not less than0.10%. |
