Objective:To observe bai zhu (Rhizoma Atractylodis Macrocephalae), fu ling (Poria) and Fu Ling Bai Zhu Tang (Rhizoma Atractylodis Macrocephalae and Poria Decoction) on the changes in the gastrointestinal tract morphology in animal models and intestinal propulsion, aquaporin (AQP), electrolyte, etc., revealed its mechanism of treating spleen deficiency diarrhea from the perspective of the water metabolism.Methods:①using spleen deficiency diarrhea model by irregular diet and breaking qi and elimination with the bitter-property medicinals.②The application of charcoal powder propelling method for the determination of rat gastric emptying and intestinal propulsive;③The electrolyte analyzer for the determination of serum electrolyte levels;④optical microscope observation of the stomach intestinal tissue morphological changes;⑤The enzyme-linked immunosorbent assay of jejunal and colonic mucosa of AQP3in of AQP4content. Comparative Study of bai zhu, fu ling and Fu Ling Bai Zhu Tang treating spleen deficiency diarrhea, and to explore its mechanism of action.Results:①The optical microscope observation showed that the treatment groups have different levels of damage to improve:the jejunum and colon parts were significantly improved in fu ling group; the rectum was significantly improved in bai zhu group and Fu Ling Bai Zhu Tang.②the rate of gastric residual were all decline, of which Fu Ling Bai Zhu Tang group was most significant (P<0.05). The rate of small intestine promote were all rise. Among them, Fu Ling Bai Zhu Tang group and bai zhu group were significant (P<0.05).③the Na+、 K+level in serum are increased to varying degrees, of which Atractylodes tuckahoe group of Na+level and the bai zhu group of K+levels were significant (P<0.05).④each group regulates expression of aquaporin:(1) of AQP3:jejunum parts are lower, bai zhu group was significant (P <0.05); colon parts are higher, each group has significant difference (P<0.05).(2) of AQP4:jejunum parts were increased, which Fu Ling Bai Zhu Tang group was significant (P<0.05); the colon area were increased, but each group was no significant difference (P>0.05).Conclusion:The mechanism of bai zhu, fu ling and Fu Ling Bai Zhu Tang treating spleen deficiency may be as follow:First, repairing the intestinal mucosal injury, improving the permeability, and promoting the absorption of liquid water. Second, regulating AQP3expression, restoring the balance of serum electrolytes, promoting intestinal inflammatory pathology exudation and permeability of solute discharge and improving the function of intestinal fluid metabolism.
