| Object:Use different Aloxidone, observe Influence and analgesic effects that rats suffering Chronic narrowing the sciatic nerve damage have on the thermal hyperalgesia and spinal cord cellsMethod:Choose weight200-250g of50Wister male rats. To adapt to the environment, raise50rats in the feed room for5days. Before operation and stomach Irrigation, do the thermal withdrawal latency, TWL.the average from the test is Hot stimulate pain threshold.According to the average, remove rats whose threshold of pain are too high or too low. After screening, choose the suitable rats for test. Choose Group A(n=10) randomly, fake opertion group:Not doing Sciatic nerve ligation, expose right sciatic nerve for3-5minutes, irrigate5ml Physiological saline; the rest rats do the the right of sciatic nerve ligation operations, establish chronic narrowing the sciatic nerve damage model, after operation, with the pain threshold, choose successful model of rats. Group B(n=10), Model control group:irrigate saline5ml In the stomach; GroupC(n=10), Carbamazepine group:In the stomach with carbamazepine6mg/kg(Soluble in5ml saline); GroupD(n=10), Gabapentin group:In the stomach with carbamazepine6mg/kg(Soluble in5ml saline); GroupE (n=10), Pregabalin group:In the stomach with pregabalin6mg/kg (Soluble in5ml saline). From after operation day, fake opertion group and Model control group irrigate daily the stomach with corresponding drugs twice Until end the test. In the1st,2nd,3rd,4th,5th,6th,7th,10th,14th day after operation, observe thermal hyperalgesia and behavioral change. The14th day, after having the rats die, take out the corresponding spinal cord segment used as model, then make pathological specimens and frozen section. Do the Immune organization to frozen section, observe the number of c-fos determination in the spinal cord neurons.Result:(1)ethology observation result:Compare Group A and4model Groups, compare their ethology changing scores in3rd days after operation, there are apparent differences(P<0.05或P<0.01); Compare GroupB and3model treatment groups, compare their ethology changing scores in3rd days after operation, there are apparent differences(P<0.05或P<0.01); Compare GroupE(14th day after operation) and GroupA, compare their ethology changing, there is no apparent difference(P>0.05);Compare GroupE, GroupC and GroupD,there are distinct differences(P<0.05).(2) Thermal pain threshold testing result:From3rd days after operation, there are distinct significance among GroupA, GroupBã€Cã€D(P<0.01), However, there are no statistics significance between GroupA and GroupE in before and after operation (P>0.05);compare GroupB and model treatment group,there is statistics significance except GroupC(P<0.05或P<0.01);compare GroupE and2model treatment Groups, there is statistics significance (P<0.05或P<0.01)(3)c-fos expression:compared with GroupA, spinal dorsal horn c-fos expressions of each model group have all incresed, they all have distinct defferences (P<0.05或P<0.01); positive rate of c-fos in GroupB is apparently higher than GroupC (P<0.05)〠GroupD (P<0.05) and GroupE (P<0.01); positive rate of c-fos in GroupE is apparently lower than GroupD, having distinct differences (P<0.05); when compared with GroupB, FLI cell numbers of each spinal cord layer in GroupE are significantly different(P<0.01); when compared with GroupD, FLI cell numbers in â… ï½žâ…¡ layers of spinal dorsal horn in GroupE, have significatly decreased, having significant differences (P<0.05).Conclusion:Carbamazepine, Gabapentin and Pregabalin have analgesic effect on the rat-Wister which has neuropathic pain. The experimental results show that the analgesic effect on the magnitude is carbamazepine<gabapentin<pregabalin, from which we can see that anti-epileptic causes spinal glial activation on peripheral nerve infection and injury, and can reduce the spinal dorsal horn neurons in afferent noxious stimulation in the process of algesthesia caused by Inflammatory factors. Among them, regulatory factors play an important part. |
