| Objective: Explore the expression of HSP27and Caspase-9and itsrelationship to apoptosis in hepatolithiasis associated intrahepaticcholangiocarcinoma. Relationship between the expression of HSP27ã€Caspase-9and the onset and progress of HLAIHCC was investigated to layan experimental base for the prevention and treatment of HLAIHCC.Materials and Methods: Sixteen cases of the tissue of bile duct fromhepatic hemangiomas or liver trauma (group A)ï¼›Forty-eight cases of thetissue of bile duct surrounding calculus of intra-hepatic(group B) andthirty-eight cases of tumor tissue of HLAIHCC(group C) were randomlysampled and collected in Hepatobiliary Surgery in Hunan ProvincialPeople’s hospital between June2006to June2010; immunohistochemistrywas employed to detect and statistically analyze the expression of HSP27and Caspase-9in each group. Tumor cell apoptosis was detected with TUNELmethod. Observe the apoptotic index and analysis the relationship betweentwo proteins and apoptosis.Result (1) In group A,B,C, the positive rate of the expression of HSP27was0ã€29.56%and73.68%,The positive rate of the expression of HSP27in the carcinoma tissue of HLAIHCC was significantly higher compared withthat in the control group(P<0.05); In group A,B,C, the positive rate ofthe expression of Caspase-9was81.25%ã€72.73%and28.95%, The positiverate of the expression of Caspase-9in the carcinoma tissue of HLAIHCCwas significantly lower compared with that in the control group(P<0.01)(2)The expression of HSP27and Caspase-9in the carcinoma tissue of HLAIHCC was not relative to the gender,age,course, tumor size and tumorpathological types(P>0.05),but their expression in HLAIHCC weresignificantly correlated with tumor differentiation degree, lymph nodemetastasis and portal vein invasion(P<0.05).(3) The expression of HSP27was followed by a low expression ofCaspase-9in the tissues of HLAIHCC. Person-related analysis show a strongnegative correlation between the expression of HSP27and that of Caspase-9in HLAIHCC (r=-0.373,P=0.021).(4)In group A,B,C, the apoptosis index(AI) were14.13±2.31%ã€13.89±4.48%and3.74±2.18%.The apoptosis index in the carcinoma tissue ofHLAIHCC was significantly lower compared with that in the controlgroup(P<0.01)。(5)The apoptosis index in patients with the positive expression ofHSP27was significantly lower than with the negative expression (4.61±3.56%Versus13.86±4.22%, P﹤0.01). The apoptosis index in patientswith the positive expression of Caspase-9was significantly higher thanwith the negative expression (14.11±3.96%Versus4.50±3.38%, P﹤0.01).(6)The apoptosis index in patients with well differentiatedã€moderately differentiated and poorly differentiated were5.50±1.69%ã€3.83±2.04%ã€2.42±1.88%。The apoptosis index of well differentiated wassignificantly higer compared with that of moderately differentiated andpoorly differentiated (P<0.05)。Conclusion (1)High expression of HSP27may be related to theoccurrence of HLAIHCC;(2) High expression of HSP27or low expression of Caspase-9may beprompted to lymph node metastasis or portal vein invasion in HLAIHCC;(3)There may be a negative correlation between the expression of HSP27and that of Caspase-9in HLAIHCC;(4)HSP27may inhibit the activity of Caspase-9to promote cell proliferation and inhibit cell apoptosis. They may play an important rolein the occurrence and development of HLAIHCC. |
