Objective: By detecting MUC3expression of MUC4in hepatolithiasis associatedintrahepatic cholangiocarcinoma, to analyze them with the clinicopathologicalparameters of intrahepatic cholangiocarcinoma, and to explore the correlation betweenMUC3and MUC4.Materials and Methods: The specimens were collected from patients in the peoplehospital of Hunan Province from June2006to June2010.One specimen is as theexperimental normal tissue of bile duct from biliary duct injury specimens, the other onefrom intrahepatic bile duct stones and the third one from hepatolithiasis associatedintrahepatic cholangiocarcinoma. Western blot is detected the expression on of MUC3and MUC4in fresh tissues; Immunohistochemical methods is detected the expression ofMUC3and MUC4in specimens of paraffin sections. SPSS17.0statistical software wasperformed to statistically analyze the relationships between clinicopathologicalparameters and their expression in HLAIHCC.Result(1) The expression of MUC3was difference in the these different Bile duct(P<0.01).Thepositive rate of MUC3protein in normal epithelial cells of bile duct is higher thanHLAIHCC (P<0.01). MUC3protein in HLAIHCC were significantly correlated withhistological grade and lymphaden metastasis(P<0.05),and they were not associatedwith age, sex, tumor size and portal Vein invasion (P>0.05).(2) The expression of MUC4was difference in the three different Bile duct(P=0.00).Thepositive rate of MUC4protein in HLAIHCC is higher than normal epithelial cells ofbile duct (P=0.00). MUC4protein in HLAIHCC were significantly correlated withhistological grade,lymphaden metastasis and portal Vein invasion(P<0.05),andthey were not associated with age, sex and tumor size (P>0.05).(3) According to a Kaplan–Meier test, MUC3positive expression showed significantly better survival than the negative expression group; MUC4positive expressionshowed significantly worse survival than the negative expression group;(4) Cox proportional hazard analysis showed that MUC3expression,MUC4expression, lymphaden metastasis and portal Vein invasion were significantindependent prognostic factors. Other factors, including age, sex, tumor size,histological grade were no significant independent prognostic factors.Conclusion(1) The protein expression of MUC3and MUC4are abnormal expression in HLAIHCC.The lower expression of MUC3and the higher of MUC4may play a catalytic role inthe occurrence of HLAIHCC.(2) MUC3expression, MUC4expression、 lymphaden metastasis and portal Veininvasion were significant independent prognostic factors in HLAIHCC, The highexpression MUC3signify a favourable prognosis;The high expression MUC4signify a poor prognosis.
