Objectives To observe the effect of renal sympathetic denervation on the blood pressure, left ventricular mass index(LVMI), renal norepinephrine and myocardial expression of TLR4, NF-κB, TNF-a, IL-6in spontaneously hypertensive rats(SHR), and discuss its possible mechanism in ameliorating left ventricular hypertrophyMethods36spontaneously hypertensive rats were randomly selected and divided into baseline group(group DO), denervation group(group D) and sham-denervation group(group S),12WKY rats with the same age were set as baseline control group(group WKY). All rats were fed in the same condition until12weeks old,then group DO and group WKY were sacrificed after recording the blood pressure.The rats in group D were bilaterally sympathectomized, the blood pressure and body weight were recorded every week. The1st week and the6th week after the denervation, group D and group S were sacrificed respectively(group Dl, S1and group D2, S2). The hearts were obtained and the ratio of the left ventricle to body weight (LV/BW) were calculated, then they were dyed with HE and Masson. The expression of TLR4,TNF-a and IL-6in the left ventricle were got by immunohistochemistry and the protein expression of TLR4,NF-κB,TNF-a, IL-6by Western blot. The kidneys were obtained to detect renal norepinephrine by HPLC-ECD.Results1.There was no difference between group WKY vs group DO, group D1vs group S1, group D2vs group S2in body weight and heart rate(p>0.05).2.The blood pressure, LVMI, NE and myocardial expression of TLR4, NF-κB,TNF-α, IL-6in group DO were significantly higher than that of group WKY(p<0.05).3.The first week after the denervation, blood pressure in group D1were significantly lower than group DO as well as group S1(P<0.05); however,6weeks after the surgery, there were no significant difference between group D2and group S2in blood pressure(P>0.05).4.LVMI, NE and the expression of TLR4, NF-κB,TNF-α, IL-6in group D1were significantly lower than group DO and S2(P<0.05); they are still lower in group D2than that of group S2(P<0.05), but close to group D0(P>0.05).Conclusions Renal sympathetic denervation can effectively lower blood pressure in a short term and significantly ameliorate LVH in SHR, and its mechanism probably mediate through restraining renal sympathetic nervous activity and decreasing myocardial expression of TLR4, NF-κB, TNF-α, IL-6.
