Objective To confirm the optimized processing technology of Fructus Arctii; To establish the fingerprints of crude drugs and processed products by HPLC; To determin multiple major components of crude drugs and processed products; To establish the qualitative model of infrared rapid quality control; To establish the IR rapid predictive model in determination of Arctiin and Arctigenin in crude and processed Fructus Arctii.Methods①Taken "Zhejiang Norms of TCM Processing"as the reference, combined with the processing history and modern literature, processed technology of Fructus Arctii was optimized. The best processing technology was gained by taking the content of Arctiin and Arctigenin, and orthogonal design experiment L9(34) used to investigate the factors of parching time, parching temperature and parching frequency, the validation experiments proved that this optimized processing technology was stable.②11batches of crude drugs were processed according to the optimum processing technology and we got11batches of processed products, the HPLC fingerprint and common pattern of11batches of crude and processed Fructus Arctii were established by using chromatographic fingerprint evaluation system.③The content of threet main components in the30batches of crude drugs and processed products were determined by HPLC.④The infrared graphs of30batches of crude drugs and30batches of processed products were collected by pellet method, and they were classified by cluster analysis of SIMCA (Soft Independent Modeling of Class Analogy), the qualitative model was establisehd between crude drugs and processed products.⑤The infrared graphs of30batches of crude drugs and30batches of processed products were collected by ATR(Attenuated Total Reflectance), and the quantitative analysis forecasting model of essential component was screened and established by combining the operation software Spectrum V6.0to the results measured by HPLC. Results①The optimized processing technology for Fructus Arctii is parching for2min at120℃, and should be flipped frequently. To authenticate the conditions mentioned above, we use the optimization craft to process three groups of Fructus Arctii and find the total count of Arctiin and Arctigenin is11.45+0.03%, it similar with the fifth text in the orthogonal experiment.②This study has established the HPLC fingerprints of crude drugs and processed products and use the software for calculating the similarity of fingerprint to set up the mode owned by both. The crude drug has12spinnacles owned by both and the processed product have19spinnacles owned by both. Then, three spinnacles of Chlorogenic Acid, Arctiin and Arctigenin have been identified. Comparing the similarities of11groups of crude drugs and processed products from different metropolis, the similarities of processed products is better than crude drugs.③A method for simultaneous determination of three major components including Chlorogenic Acid, Arctiin and Arctigenin is established by high performance liquid chromatography with diode array detector and it proves to be safe and stable.④This study has established a qualitative model by cluster analysis combined with infrared spectra and SIMCA (Soft Independent Modeling of Class Analogy), so as to classify the crude drugs and processed products, the clustering effect is excellent and the accuracy of recognizing unknown sample is over90%.⑤This study has successfully established a model to predict the content of harpagoside which is the major component in crude drugs and processed proudcts by ATR(Attenuated Total Reflectance) detection technique. The prediction results of Arctiin and Arctigenin are excellent according to the predictive model of crude drugs and processed proudcts.Conclusion①The optimizing processing technology is stabile and reliable, it could guide the production and act as a model for processing standardization.②The fingerprint could be applied to the intrinsic quality control of crude and processed Fructus Arctii.③The research of major components in crude and processed Fructus Arctii by HPLC provided a new method and idea for the further study.④The infrared qualitative model of crude and processed Fructus Arctii could provide a rapid-test for Fructus Arctii, ensure the real time detection of drug and identify their quality.⑤The infrared quantitative model of crude and processed Fructus Arctii could provide a rapid-test of the essential components in Fructus Arctii, greatly avoid time and labor consuming of the former method and the "Rapid and none pulse" advantage of infrared spectrum.
