Fluorescence quantum yield of arctiin was measured to be 0.084 at maximum excitation wavelength 280 nm.Influences of the solution acidity,surface active agent SDS and SLS on fluorescence spectra were investigated.It was found that the fluorescence spectra of arctiin and arctigenin were the same,but the fluorescence intensity of arctiin was the strongest at alkalic condition and was not influenced by coexisting arctigenin at this condition.Based on the spectral characteristics,a new fluorescence method was proposed for the analysis of arctiin in Fructus Arctii.The detection limit of method was 0.040μg ml-1.The content of arctiin in Fructus Arctii was determind to be 6.02%and 6.09%by using the standard curve method and standard addition method.According to fluorescence properties,a new thin-layer fluorescence scanning method for analysis of arctiin in Fructus Arctii was established.The results showed that the relationship between chromatographic peak area and arctiin quality was linear over the range of 0.270μg to 1.35μg(r=0.999),the recovery rate was 98.2%. The precision,stability and reproducibility of the method were fine.2.Dauricine and daurisoline extracted from Rhizoma Menispermi were identified by nuclear magnetic resonance(NMR) and mass spectrum(MS).Spectral properties of Rhizoma Menispermi and four of its active compounds:dauricine,daurisoline,tetrandrine and sinomenine,were researched.Influences of the solution acidity on fluorescence spectra were investigated.A new thin-layer fluorescence scanning method for analysis of tetrandrine in Rhizoma Menispermi was established.The results showed that the relationship between chromatographic peak area and tetrandrine quality was linear over the range of 0.248μg to 1.24μg(r=0.999),the recovery rate was 97.1%.The precision,stability and reproducibility of the method were satisfactory. | | Keywords/Search Tags: | Traditional Chinese medicine, Fluorescence analysis, Thin-layer fluorescence scanning method, Arctiin, Arctigenin, Dauricine, Daurisoline, Tetrandrine, Sinomenine | PDF Full Text Request | Related items |
