BackgroundSince the1930s,Sodium pentachlorophenol was used as a disinfectant,herbicide andwood preservative in the global range widely used. Sodium pentachlorophenol ’s main purpose isto oncomelania elimination and schistosomiasis prevention. Sodium pentachlorophenol couldbioamplificate through food chain, thus leading to different levels of biological damage.Sodium pentachlorophenol through the biology byskin, sodium respiratory tract, enteron andother ways. Organisms are affected through system biology toxicity, reproductive developmenttoxicity, endocrine disturbance, and cancer, the influence of immune toxicity.pentachlorophenol and pentachlorophenol sodium affects reproductive development with weakestrogen function and enzyme function change. They interfere with the normal function of thethyroid element. Sodium pentachlorophenol itself is not toxic, but their metabolic product istoxic. To develop a method of detecting Sodium pentachlorophenol in blood plasma. To fullyunderstand the Sodium pentachlorophenol metabolic process has the vital significance.ObjectiveTrying to use the capillary chromatographic column DB-35MS, exploring the appropriatecondition for the chromatographic analysis of sodium pentachlorophenol. A method ofpentachlorophenol sodium in blood sample by the capillary gas chromatographic simultaneouslywas developed and validated, so as to further improve the determination method forpentachlorophenol sodium in blood sample—the occupational health standards GBZ/T210.5-2008, which could better satisfy actual work.MethodsThis method to establish using sulfuric acid acidification, pentachlorophenol sodium wasacidification into pentachlorophenol. The samples were extracted by n-hexane, Potassiumcarbonate stripping, acetic anhydride derivative, and the characters of gas chromatography(GC)method were studied.1.Create blood sample pretreatment methods built in pentachlorophenol sodium;2. Determined by gas chromatography parameters in pentachlorophenol sodium; 3. The labeled recovery is determined;4. The lowest concentration determination of detection.;5. Analyze the stability of blood the in pentachlorophenol sodium content.6.Precision experiment research.ResultsThere was a good linearity in a range from2.5μg/kg to500μg/kg, with correlationcoefficients (r) of0.9992and linear regression equation for toluene was Y=136.45X-134.26.,The detection of limit (LOD) of the method was0.3μg/kg. The average recovery rate rangedfrom76.7%to115.0%.,relative standard deviation (RSD) less than10%.ConclusionsThe method was found to have higher sensitivity with the minimum detectionconcentration of toluene for0.3μg/kg after practical application for determination ofpentachlorophenol sodium in blood sample, far below1.0μg/kg, the recommended minimumdetection concentration of pentachlorophenol sodium in blood sample, namely T3030SC/T in2006-determination of pentachlorophenol and its sodium salt residues in fishery Gaschromatography method. What’s more, the method satisfied requirements of determinationmethods of chemicals in biological materials(GBZ/T210.5-2008). Capillary column DB-35MSused was selective, and the separating effect was very good, with the optimization of peakshape. It can completely separated from the blood of pentachlorophenol acetate (PCP-Oac).
