Expression Of COX-2and MMP-2in Breast Cancer And Their Clinical Signigcance

Objective:To assess the expression of COX-2and MMP-2in human breast cancer, breastfibroadenoma, normal breast tissue respectively,and analyze their correlation inbreast cancer tissue, Further discusses the breast cancer occurrence, development,Invasion and metastasis of the function and the clinical significance.Methods:Collect82cases of paraffin blocks who had selected at pathologists in nanch-ang university of the first affiliated hospital randomly in2010-2011for study.andwith its as the research object. Among them,50cases definitely diagnosed as prim-ary breast cancer specimens(all are ductal carcinoma),17cases as fibroadenoma ofthe breast and15cases as normal breast tissue(from the margins of breast cancer atleast5cm). All case of illness have complete clinical data, for first surgery, beforewhich without any anti-tumor treatment.Detect COX-2and MMP-2expres-sionlevel with the EnVision immunohistochemistry,and compares with the clinicalparameter．Results:1、The positive rate in breast cancer about COX-2is60％,24％in breastfibroadenoma, and13％in normal breast tissue.COX-2of positive breast cancer wassignificantly higher than the breast fibroadenoma and normal breast tissue, which aresignificant deviation(p<0.05)．2、The expression of COX-2is negative correlated to the pathologic and TNMstage of tumors(r=-0.383,-0.356,respectively). When lympho node matastasis andVascular infringement occurs, the more lympho nodes metastasis and Vascular infrin-gement, the more COX-2expressed(r=0.344,0.414,respectively).But the expressionof COX-2do not correlated with the ege and the size of the breast cancer patients(p＞0.05). 3、The positive rate in breast cancer about MMP-2is68％,35％in breastfibroadenoma, and13％in normal breast tissue. MMP-2of positive breast cancerwas significantly higher than the breast fibroadenoma and normal breast tissue, whichare significant deviation(p<0.05).4、The expression of MMP-2is negative correlated to the TNM stage oftumors(r=-0.356). When lympho node matastasis and Vascular infringement occurs,the more lymphonodes metastasis and Vascular infringement, the more MMP-2exp-reed(r=0.461,0.492,respectively).But the expression of MMP-2do not correlated withthe ege, the size and the Pathology classification of the breast cancer patients(p＞0.05).5、There was positive correlation between COX-2and MMP-2protein express-ion in the breast cancer (P＜0.05, r=0.490).Conclusions:1、COX-2and MMP-2of positive breast cancer was significantly higher than thebreast fibroadenoma and normal breast tissue.2、The high expressions of COX-2and MMP-2has the close relationship to theInvasion and metabasis of breast cancer．3、The combination detection of COX-2and MMP-2have the positive referenceof the judge of malignant phenotype. Can evaluate the prognosis of patients withbreast cancer and provide more the theory reference for targeted therapy.
