Experimental Study About The Intervention Of Yixintai On Plasma Of RAAS In Rats With Chronic Heart Failure

Purpose:This study was designed to identify the effect of YIXIN Thai Traditional Chinese Medicine Pill on ventricular remodeling in rats with coronary heart disease. And illustrating the mechanism by associated indicators of renin-angiotensin-aldosterone system(RAAS), proinflammatory cytokines, and signal transduction pathways.Method:We had selected the Wistar healthy male rat whose weight is Between200gand250g, the use of abdominal aorta Act set up chronic congestive heart failure animal model. The success of the model rats were randomly divided into five groups:group modulus space, high-dose group of Chinese medicine, Chinese medicine in the dose group, Chinese medicine, low-dose group and western medicine,10in each group; separate a group of10normal control group. Animals in each group of more than four weeks after administration, respectively (in addition to the normal control group) using ELISA method, ABC immunohistochemical method were determined in plasma PRA, Ang II, ALD and myocardial tissue AT1receptor protein content.Result:Normal group and model group is obviously different, the general state of Model rats is poor (less activities, dull hair, cyanosis fo lips and terminal claws). Concentration of plasma PRA, Ang II, ALD, TNF-aand IL-6is far higher than the normal group (P <0.01); while YIXIN Thai group, Catopril group and Compound Salvia pellet group is lower compared with model group(P <0.01).at the same time,the amout of PKC in rats myocaidial tissue in model group is higher than sham-operation group(P<0.01) and YIXIN Thai group, Catopril group and Compound Salvia pellet group is lower compared with model group(p<0.05) which proved this signal transduction substance participates in ventricular remodeling.Conclusion:1.YIXIN Thai pills can reduce the content of PRA、 Ang II、 ALD、 TNF-α、 IL-6in plasma. It may interference ventricular remodeling by inhibiting the activation of neuroendocrine system and regulating inflammation cytokine.2. YIXIN Thai pills can reduce the content of PKC. it may supress ventricular remodrling by regulating the activation of PKC in myocaidial tissue.
