Objective: To investigate the expression and correlation of TRPV6,CCK-B and β-catenin in colon polyp/carcinoma tissues by detecting the protein and mRNA expression of TRPV6,Gastrin/CCK-B and β-catenin during colon polyp canceration.Methods: Real-time PCR was used to detect the expression of CCK-B,TRPV6 and β-catenin mRNA in normal colon mucosa tissue,colon hyperplastic polyp tissue,colon adenomatous polyp tissue and early colon cancer tissue samples.Western blot and immunohistochemistry were used to detect the expression levels of Gastrin/CCK-B,TRPV6 and β-catenin proteins in the above-mentioned colon tissues,and the expression and correlation of appellate indexes were analyzed.Results:(1)Immunohistochemical results showed that the expression intensity of β-catenin,Gastrin and TRPV6 in normal colonic mucosa,proliferative colonic polyps,adenomatous colonic polyps and early colon cancer tissues had a trend of gradual increase,and the difference was statistically significant(P <0.05).(2)Real-time PCR results showed that TRPV6,CCK-B and β-catenin mRNA were expressed in normal colonic mucosal tissues,and β-catenin expression was gradually up-regulated in proliferative polyps of colon,adenomatous polyps of colon and early colon cancer tissues,and significantly increased in early colon cancer tissues(P <0.05).The mRNA expression of TRPV6 and CCK-B in colorectal adenomatous polyps was significantly higher than that in colonic hyperplastic polyps(P <0.05),and the mRNA expression of TRPV6 and CCK-B in early colorectal cancer was significantly higher than that in colorectal adenomatous polyps,but the difference was not statistically significant(P >0.05).(3)Western blot results showed that TRPV6,CCK-B and β-catenin were also slightly expressed in normal colonic mucosal tissues,and the expression of TRPV6,CCK-B and β-catenin in proliferative colonic polyps,adenomatous colonic polyps and early colon cancer tissues were gradually up-regulated,with statistically significant differences(P <0.05).(4)By Western blot test normal colonic mucosa tissue,colonic hyperplastic polyp,colonic adenoma polyp tissue,early signs of colon cancer tissues TRPV6,CCK-B,β-catenin protein expression,the use of Spearman correlation between analysis indicators,the results show that the beta-catenin expression level was positively related with CCK-B(r = 0.988,P = 0.012),and TRPV6 expression were positively correlated(r = 0.998,P = 0.002),and the differences were statistically significant.Conclusion:1.The expression of Gastrin/CCK-B,TRPV6 and β-catenin proteins and mRNA in normal colonic mucosa tissues,proliferative colonic polyps,adenomatous colonic polyps and early colon cancer tissues increased gradually.2.CCK-B and TRPV6 proteins were positively correlated with β-catenin in normal colonic mucosa,proliferative colonic polyps,adenomatous colonic polyps and early colon cancer tissues. |
