Obesity is increasingly becoming a globle public health problem. Once associated with high-income countries, obesity is now also prevalent in low- and middle-income countries.Obesity is the most common cause of insulin resistance and type 2 diabetes.Poorly controlled diabetes over the long term tends to cause heart,brain, kidney, nerve, eye, and other organ damage. The prevalence rate of diabetes which has become a major public health problem in China,has reached 9.7 percent.The essay was to choose three compounds(including Huashi Compound,Qingre Compound and Wenyang Compound),which were based on Dr. Chen analysis of a combination of clinical manifestations,etiology and patheogenesis of obesity and diabetes. The major were to observe the effects of various compounds on glycolipid metabolism in obese rats and diabetic rats, sceen the effective compound.Then the study was to explore its possible mechanism,thus providing experimental evidence for the prevention and treatment of metabolic abnormalities,which are associated with obesity and diabetes.Firstly,the screening for the effective compound for the treatment of obesity has been conducted in obese rats. The rats were fed ad libitum during 3 months on a high fat diet (HFD)to induce obesity.The effects of various compounds on glycolipid metabolism and pathological changes in obese rats were observed.Results showed that three compound could inhibit blood sugar, reduce serum triglycerides, total cholesterol, liver triglyceride and free fatty acids, reduce the extent of tissue lesions.Particularly, Wenyang Compound performed better.Then the levels of serum glycated hemoglobin (GHbAlc), insulin (INS), tumor necrosis factor-α(TNF-α), endotoxin (ET) and cyclic nucleotides in Wenyang group were detected.The results showed Wenyang Compound could reduce the leves of serum GHbAlc,INS,TNF-αand ET,increas the proportion of cyclic nucleotides.This indicated that Wenyang Compound was an effctive drug in treating obesity, thus lowering lipid and blood glucose, partially improving insulin sensitivity, intestinal microflora, anti-inflammation and the body’s basal metabolism. Secondly,The compound screening was performed in diabetes rats.We established non-obese diabetic rats with a combination of high-fat diet-fed and low-dose streptozotocin. The effects of various compounds on glycolipid metabolism and pathological changes in diabetic rats were observed. Results showed that Qingre Compound could inhibit the blood sugar, improve glucose tolerance, decrease liver TG, FFA levels, reduce the heart, liver and pancreas tissue damage,but Huashi Compound and Wenyang Compound performed poor in lowering blood sugar.Then the levels of serum INS,TNF-α,ET and cyclic nucleotides in Qingre group were detected.The results showed that Qingre Compound could increase INS level,reduce TNF-αcontent,and adjust the ratio of cyclic nucleotides.Obeviously,Qingre Compound for antidiabetes had prominent effects include reduction in blood sugar,liver TG and FFA.To a certain extent,Qingre could improve glucose tolerance and insulin resistance, inhibition of inflammatory factors and reduce organ damage.As described above, these two experiments prove that three compounds could partially improve the disorders of glucolipid metabolism.Results indicate that traditional Chinese medicine(TCM) treatment of obesity is focused on strengthing kidney-yang, while TCM treatment of diabetes is more prone to clear away heat and eliminate toxins. |
