Medicine Science are closely related to human, and human disease has become a increasing concerned problem. Research and Utilization of disease knowledge has aroused high attention and concerned with the application and development of the Internet and the information technology,. However, the current knowledge management system has some problems ,on the one hand,the traditional method of medical knowledge organization are often limited to a certain professional scope,separating the interrelated medical knowledge which can obstruct the acquisition and utilization of the full and accurate medical knowledge by people. On the other hand,the current participation in information retrieval is the external form of the character, rather than their concept,and lack the uniform semantic description of information resources, so the computer can not correctly understand the semantic content of medical knowledge. And the computer can not guarantee the accuracy of the content matches, therefore the search results often return a large number of useless information, causing the user can not accurately and quickly find the resources they need.Ontology provides a new way to solve these problems as a conceptual modeling tools which can describe the semantic medical information. Ontology provides a common understanding of domain knowledge, and presents clear definition of these domain knowledge concepts and the semantic relation between these concepts from different levels of formal models.Ontology can provide the acquisition and sharing of the medical information in semantic formal, and mine large amount of the hidden information and knowledge in the network information resources. It can solve the problem of information organization, and better meet the needs of users who retrieval medical information.Firstly,this paper widely analyzes the research content and status of domestic and international domain ontology, summarizes the problems with the current theoretical research and practical exploration of ontology, proposes knowledge reconstruction by changing the tree structure into the network structure and a brief description of knowledge to deep description of knowledge which they can deeply reveal the semantic relationships and contents characteristics of the disease knowledge, making it easily to acquire disease knowledge and information. This paper proposes to build ontology stagely.First it build the core ontology, then one by one add related concepts and relationships of the field to the core ontology one by one to extend the existing ontology,so make the plans of ontology construction more controllability ,expansibility and feasibility.Secondly,this paper describes the basic theory of ontology and analyzes the advantages of organizing disease knowledge based ontology.Then this paper comparatively analyzes the current major ontology language, ontology construction tools and the method of ontology construction, then finally select the OWL as disease domian description language and Protégéas a disease domian construction tool, then this paper discusses the method of constructing domain ontology according to seven-step method.Finally, this paper definites the core concepts of disease knowledge, the properties of the core concepts,semantic relationships and domain knowledge axiom, discusses the method of constructing domain ontology according to seven-step method, and constructs the Disease Domain Ontology according to the method of building disease ontology by the Protégé,and sets up the initial ontology model of Disease Domain which can organize the medical resources orderly and use it efficiently and provides some methods and examples for establishing practical system.
