CIRI refers to that brain cell damages deteriorate when blood is poured into after cerebral ischemia, which includes primary damages, secondary ones with blood poured into. The blood-poured into damages are caused by the combination in many elements.Study shows that the regeneration of blood vessels decides ischemic neuron'survival and it is critical for making damages better to recover blood flow volume in half-gray parts which are short of blood. New-born blood vessels are made by VEGF which is the only source producing mitosis in vascular endothelium and encourages new blood vessels to develop, improves the circulation of lateral branches, inhibits the death of neve cells.The study evaluates the damages of nerval functions, measure the volumes of cerebral infarction of targeted rats at different time, determine HE chromosome and VEGF in rats, nidogen antibody, the intensity of immunism fluorescence in positive cells, then discuss Guanxin shutong's protective function and then provides the theoretical support in clinical treatment.Object i ve:Through thread embolism method,make rat'cerebral ischemia damages when blood poured into,measures the volumes of cerebral infarction of targeted rats at different time, determines HE chromosome and VEGF of homogeneous syrup in brain tissue, nidogen antibody, the intensity of immunism fluorescence and discusses Guanxin shutong' s protective function in the treatment of rat'cerebral ischemia damages when blood poured into.Method:Use male healthy Wistar rats,randomly group them into four:one with high dose of Guanxin shutong,another with low dose,and ischemia group with blood poured into and sham group.Each group has ten rats.Make rats short of blood 120 minutes than input 6h,12h,24h,48h and 72h.Evaluate their neural function damages, then by 2,3,5TTC dyed, and measure the volumes of cerebral infarction;dying HE,homogenateVEGF and Nestin antibody, inspect the changes of damaged cells.With schemia rats with blood poured into by 3d,7d, determine VEGF; With the immunism fluorescence of Nestin antibody, determine the intensity of positive cells..Result: Nerval functions of rats with high doses are recovered obivously after modeled. The high-dose group(6h,12h,24h) has obvious reduction in the volume of cerebral infaction compared with the model group (p<0.05); so does the high dose group of 48h; in these groups, the 24 h group is the most outstanding (p<0.05) The group of ventricle HE dyed has no lestion but the group of ischemia-reperfusion injury tend to worsen with the length of time. In the low dose group, it is 72h>48h>24h> 12h>6h, and the tendancy of lestion is 6h<12h<or=24h<48h<72h. Cells in high dose group changes better than the low one and the group of ischemia -reperfusion injury. The 24 h in high dose group is close to be normal. In the VEGF immunism coloration, the following of 6h,12h,24h and 18h has no effect on cortical neurons but they have effect on white neurons. Both the high dose and low dose in 12h and 24 h have obvious effect but the 72h high dose hasn't obvious effect. The low dose of 48h and 72h, ang the high dose of 12h,42h,48h,72h have evident effect on blood capillary. In the ventricle immunism coloration group, both of 7h and 12h in high dose have evident effect on cortex. In the sham group, vascular endothelium growth factor in rats'brain homogenate has expression: injected for 7 days, the high dose of 7d, the low dose have obvious expression in comparison with the model group(p<0.05). VEGF in cerebral ischemia-reperfusion injury ascends in contrast with the sham group but the 3d high dose group and the low dose one have no obvious changes. In the contrast of fluorescence intensity, the Cerebral ischemia-reperfusion injury group (3d), the model group's slant cort damaged and SVZ have Nestin.With blood poured into, the 7d group has outstandingly positive cells but the high dose doesn't.the group of 7d has outstanding changes in the number and form of Nestin positive cells..Conclusion:Guanxin shutong has protective effect on the damages of focal cerebral ischemia of rats when blood poured into.
