Objective:To observe the effect of Guanxin Shutong capsule on the expression of tissue morphology,blood lipids,blood flow and inducible nitric oxide synthase(iNOS)in early aortic atherosclerosis(AS)rats,and to elucidate the mechanism of cardiac shutong capsule on early as rats,and to provide experimental basis for traditional Chinese medicine in preventing and controlling as diseases.Methods:Sixty healthy male rats were randomly divided into five groups: blank control group(con),model group(M),atorvastatin calcium group(A),low-dose Guanxinshutong capsule group(DY)and high-dose Guanxinshutong capsule(GY)group,with 12 rats in each group.The blank control group was fed with common basic diet,while the other four groups were fed with high-fat diet for AS modeling.The model group and the blank control group were fed with the same amount of saline,atorvastatin calcium tablets were fed with atorvastatin calcium(10mg/kg/d),high-dose Guanxin Shutong capsule(GY)was fed with(1.8g/Kg/d)and low-dose Guanxin Shutong capsule(DY).Gavage(0.6g/kg/d)was given once a day for 9 weeks.At the end of 12 weeks,serum lipid level,whole blood viscosity,hematoxylin(HE)staining and iNOS expression were measured.Results:1.General conditionsThe weight of rats in model group increased significantly and activity decreased,while the weight of rats in atorvastatin calcium group,high and low dose Guanxinshutong capsule group was lower than that in model group.Compared with the atorvastatin calcium tablet group,there was no significant difference in body weight between the high and low dose Guanxinshutong capsule group.In the course of intervention,the observation found that the model group rats were depressed and their hair color was dim.Compared with the model group,the changes of the above indexes were slight,especially in the high dose Guanxinshutong capsule group.2.Blood lipid levelsCompared with the blank control group,the levels of TG,TC and LDL in the model group increased significantly,while the levels of HDL decreased significantly(P < 0.05).Compared with the model group,the levels of TG,TC and LDL in the drug intervention group decreased significantly,while the levels of HDL increased significantly(P < 0.05).Compared with atorvastatin calcium tablet group,TC in high dose Guanxin Shutong capsule group increased significantly(P < 0.05),while TG,HDL and LDL had no significant difference(P > 0.05);TC and LDL in low dose Guanxin Shutong capsule group increased and HDL decreased(P < 0.05),but there was no significant difference between TG and atorvastatin calcium tablet group(P > 0.05).3.Changes of whole blood viscosityCompared with the blank control group,the whole blood viscosity of the model group increased significantly(all P < 0.05);compared with the model group,the whole blood viscosity of the drug intervention group decreased significantly(all P < 0.05).The whole blood viscosity of high dose Guanxin Shutong capsule group was lower than that of atorvastatin calcium tablet group(P < 0.05);there was no difference between low dose Guanxin Shutong capsule group and atorvastatin calcium tablet group(P > 0.05).4.Morphological observation of aortaUnder light microscope,the intima and intima of aorta were thickened,inflammatory cells infiltrated,fibroblasts proliferated in the outer layer,and the structure was disordered and incomplete in the model group;in atorvastatin calcium tablet group,pathological changes of intima and intima were not obvious,and the outer structure was not clear;in high dose Guanxinshutong capsule group,the intima was slightly thickened,a small amount of fibroblasts proliferated in the middle layer,and the outer structure was relatively intact;In the dosage Guanxinshutong capsule group,the intima was slightly thicker,there was a small amount of fibrous tissue proliferation in the middle layer,and the outer structure was incomplete.5.The iNOS expressionCompared with the blank control group,the expression of iNOS(OD)in the model group was significantly increased(P < 0.01).Compared with the model group,the OD value of each intervention group was significantly lower(allP < 0.01).Compared with atorvastatin calcium tablet group,the OD value of high dose Guanxin Shutong capsule group had no significant difference(P > 0.05).Compared with atorvastatin calcium tablet group,the OD value of low dose Guanxin Shutong capsule group had significant difference(P < 0.05).Conclusion:Guanxinshutong Capsule can regulate lipid metabolism,reduce blood viscosity,improve aortic pathological morphology and reduce iNOS expression in early atherosclerotic rats,and play an antagonistic role in the further development of AS. |
