Effect Of Shexiang-wulong Pill On Morphology And Expression Of OPG, RANKL In Cartiage Of Rats With Adjuvant Arthritis

Objectives To observe the effects of shexiang-wulong(SXWLW)pill on Pathom-orphology and expression of receptor activator of nuclear factor-kB ligand (RANKL) and Osteoporotegerin (OPG) of cartiage in rats with adjuvant arthritis.Methods 1.Divide the rats into 3 groups and establish model: number the 30 Wistar rats (female and male was equal) according to their weight and divide them at random into3 equal groups: Group A: normal comparison group; Group B:adjuvant arthritis group; Group C: Musk-Aconite-Earth- worm Pill group. To set model of AA, the rats in group B and C were endodermicly injected Freund's complete adjuvant 0.1ml into voix pedis of right postpedes.2.Provide medicine: At the 15th day after making model,each rat of group C was intragastricly administrated MAEP (dissolved in 2ml normal saline) 7 days, according to its equivalent dosage/㎏/day(10 times of person's dosage /kg/day), 1/day; group A and B were given 2ml normal saline in the same way.3.HE dyes, observe the joint pathological change under the light microsco- Pe and assess pathological score, and detected the expression levels of OPG and RANKL collagen in cartilage tissue by Cartilage tissue section immunohistochemistry.Results 1.Observe the changes of pathomorphology by light micro scope: Group A the structure of cartilage of joins are intact without coloboma ,car- tilage cell arranged orderly.Group B quantity of cartilage cell decreased obviously ,arranged disorderly, there was obvious ulcer in cartilage, degra- dation and fibrosis. Group C the syndromes appeared in Model group were reduced significantly.The pathological score: group A 1.3±0.483,group B 3.5±0.527,group C 2.9±0.568. group B and group C has the obious difference compared with group A (P<0.01). group C has the obious difference compared with group B (P<0.05).2 Effects on the immunohistochemistry result of OPG and RANKL expression in cartilage tissue: The expression of OPG is higher than model group and normal group,and the expression of RANKL is Lower than model group.Conclusion SXWLW could reduce the ratio of OPG/RANKL to inhibit the activation of osteoclasts,therefore it could decrease cartilage damage.
