The Effect Of Musk-Aconite-Earthworm-pill On The Pathomorphology And VEGF, ES Of Synovium In The Rats With Adjuvant Arthritis | | Posted on:2012-03-29 | Degree:Master | Type:Thesis | | Country:China | Candidate:C J Ding | Full Text:PDF | | GTID:2214330368475020 | Subject:Chinese medical science | | Abstract/Summary: | PDF Full Text Request | | Objectives To observe the effects of Musk-Aconite-Earthworm-pill on pathomorphology and expression of vascular endothelial growth factor (VEGF) and endostatin (ES) of synovium tissue in the rats with adjuvant arthritis (AA), and investigate Musk-Aconite-Earthworm-pill treats the rheumatoid arthritis whether concerns with the adjustment to the imbalance of VEGF/ES.Methods1.Divide group and build model: The Wistar rats in the experiment are 30 in all and about 180 grams(20 grams more or less) .They are numbered according to weight from large to small and from 1 to 30.Then the rats is divided into three groups randomly and averagely.The groups are normal controls group (A) ,model group and Musk-Aconite-Earthworm-pill group and one has 10 rats.Freund's complete adjuvant is injected to the rats' right hind feet by 0.1 ml each in order to establish AA rats' models with A and B groups.2.Give medicine: Irrigating stomach begins the 15th day of mading mode. In C group ,Musk-Aconite-Earthworm-pills are taken by the rat with 10 times(1g/kg) of the adult one time a day. Also the boluses are dissolved in the physiological saline by 1g/2ml. The A and B groups take the same quantity physiological saline.All the groups are irrigated by a week. Of course during the week,rats are raised conventionally.3.Observe the rats in synovial histopathologic morphology with light microscope.And calculate the integral of pathology according to the result.4.Detect the VEGF and ES in the metatarsophalangeal joint of the rats with the immunohistochemical method.Results1.General situation observed:In the experimental process in A group,the rats were in a good state.Their furs were luster and they actived freely.In B and C group during 1th-3th days the rats showed acute inflammatory response and their right hind feet showed red and swollen obviously.Meanwhile their spirits were flagging and the furs were messy with less food and activities. During 12th-14th days,they showed secondary pathological changes in the tails,ears,fores and left hind feet what showed red and swollen, nodules and limbs actived obstacles.In C group,the rats' pathology changes reduced gradualy,ate more,activted more and the swelling feet changed better.2.Observe the changes of synovial histopathologic morphology in light microscope: In A group, the synovial organization surface was smooth , looked like thin film, seemed translucent. And the synovial cell is of monolayer, aligned and flat. Here was no vascular proliferation, no fibrosis and no inflammatory cells infiltrating; In B group, the synovial organization surface was rough, swelling obviously, hyperemia of and hypertrophy. The synovial cells were layers of and hyperplasia lively. In synovial organization, here were more fibrous tissues.Blood capillaries were expansive, hyperemia of, much inflammatory cells and fiber mother cells infiltrating;In C group, the synovial was less swelling and hyperemia of than it in B group. Here, the synovial organization was hyperplasia lightly and slightly inflammatory cells infiltrating.The vascular in it was proliferation of and the fibrosis was not obvious. The pathological integral in B and C group were higher than it in A group (p<0.01). The pathological integral between C and B group is different obviously (p<0.05).3.The masculine expression of VEGF and ES: The masculine exprssions of VEGF and ES were fixed in cytoplasm and cell membrane and seemed yellow or tan partcleses.They mainly distributed in synovial innermost layer and lower levels. VEGF masculine expression in A group was higher than it in B group(p<0.01). And it was lower much in C group than in B group. ES was no expression in A group and low expression in B group. It was higher much in C group than in B group(p<0.01).4.The ratio of VEGF/ES: The ratio of the VEGF and ES was much lower in C group than in B group(p<0.01).Conclusions The experiment proved that Musk-Aconite-Earthworm-pill could improved synovial organization's pathological degree of damage with the model of the rats whose metatarsophalangeal joint cought rheumatoid arthritis, inhibited synovial inflammation effectively, reduced edgradation of the joint-cartilage-matrix.
