The Effect Of Musk-aconite-earthworm Pill On Pathomorphology And Expression Of BFGF TGF-β1 In Synovial Lining Layer Of AA Rats

Objective To observe the influences of Musk—Aconite—Earthworm Pill on pathomorphology and expression of Basic fibroblast growth factor (bFGF) and Transforming growth factorβ1(TGF—β1) in synovial lining layer of rats with adjuvant arthritis(AA),and to explore its therapeutic effectsrats on rheumatoid arthritis (RA).Methods 30 healthy Wistar rats were randomly divided into three groups: normal group(A group), model group(B group), and Musk—Aconite—Earthworm pill group(C group). To establish the rats with adjuvant arthritis model using Complete Freund adjuvant, fills thestomach with Musk—Aconite—Earthworm pill one week. Then, pathomorphological changes of synovial lining layer, synovial lining cell layers and expression of bFGF and TGF-β1 were observed .Results1.The observation of synovial lining layer pathological form: Group A : rats'synovial lining layer building up by 1-2 layer flat type or low set square epithelial appearance of synovial cells and some of discontinuous , synovial organization without hypertrophy and congestion. Group B: hyperplasia of the synovial lining to 4 to 8 layers in sample or the fence papillary layer sample swelled ,and the cells are ovoid, arranging disorderly, increasing in size; Synovial organization is hypertrophy, congestion and oedema, forming many fluff, quilted axon to joints, or invades cartilage and bone. The lining layer in group C belows it in group B, synovial cells mild hyperplasia, synovial organization hyperemia edema ease.2. Result comparison of cell layers of Synovial lining layer and bFGF, TGF-β1 determination:2.1 Synovial lining layers in group B and C compared with group A, layers below in group A, a statistically significant difference (P < 0.01); Group C compared with group B, layer below the B group, a statistically significant difference (P < 0.05).Experimental results show that Musk-Aconite-Earthworm pill can reduce cell layers of synovial lining layer, and synovial cell hyperplasia .2.2 bFGF expression level in group B and C compared with group A , bFGF expression above group A ,a statistically significant difference (P < 0.01); Group C and group B compared , the expression of bFGF belows it in group B, a statistically significant difference (P < 0.05); Experimental results show that Musk-Aconite-Earthworm pill can reduce the bFGF expression level of the synovial lining layer .2.3 The expression level of bFGF, TGF-β1in group B and C compared with group A, the expression of bFGF, TGF-β1 aboves in group A, A statistically significant difference (P < 0.01); Group C compared with Group B ,the expression of TGF-β1 belows it in group B , it is a statistically significant difference (P < 0.05).Conclusion(s) Experimental results show that AA rats fills the stomach with Musk-Aconite-Earthworm pill one week, Musk-Aconite-Earthworm pill can significantly reduce hyperemia ,edema and the hyperplasia of synovial membrane cell, reduce lining layer cell layers ,also significantly reduce lining layer expression level of bFGF, TGF-β1 .Clew that the treatment mechanism of Musk-Aconite-Earthworm pill in RA may reduce lining layer expression level of bFGF, TGF-β1 , Inhibit synovial lining layer cell hyperplasia, reduce blood vessels′formation, thus achieves the function to protect of cartilage and bone...
