"Banloubeilianjigongwu"is an important principle of Chinese medicine Combination Taboo theory i.e. Aconitum(Radix Aconiti,Radix Aconiti Laterlis Preparata,Radix Aconiti Kusnezoffii) could not be combined with Rhizoma Pinelliae,Fructus Trichosanthis,Bulbus Fritillariae,Radix Ampelopsis and Rhizoma Bletillae,the co-decoction of Radix Aconiti Laterlis Preparata with others may be highly toxic,however,few scholars believe that the compatibilities mentioned above are appropriate. The components of Traditional Chinese Medicine are very complicated,both a comprehensive understanding of the chemical components and the identifying the key factors of the poisoning mechanism are very necessary.In this thesis,the Aconiti Laterlis Radix Preparata was selected as the target. To investigate the Radix Aconiti Laterlis Preparata with Rhizoma Pinelliae,Fructus Trichosanthis,Bulbus Fritillariae,Radix Ampelopsis and Rhizoma Bletillae,by determining the contents of aconitine in the water extracts by boiling separately and after combination; isolation and purification the component from the Radix Aconiti Kusnezoffii;study on the biotransformation of monoester-diterpenoid alkaloids of the metabolite of aconitine in rats intestinal bacteria.The HPLC and ESI-MS method for the determination the content changes of diester-diterpenoid in preparations of aconitine compounded before and after with Rhizoma Pinelliae,Fructus Trichosanthis,Bulbus Fritillariae,Radix Ampelopsis and Rhizoma Bletillae. The content of diester-diterpenoids (aconitine,mesaconitine,hypaconitine) in decoction of aconitine compounded with Rhizoma Pinelliae, Fructus Trichosanthis, Pericarpium Trichosanthis, semen Trichosanthis,Bulbus Fritillariae Thunbergli,Radix Ampelopsis and Rhizoma Bletillae, respectively, the contents of diester-diterpenoid alkaloids are more than that of single Radix Aconiti Laterlis Preparata decoction. For the decotions of Radix Aconiti Laterlis Preparata combined with Rhizoma Pinelliae Preparata,Bulbus Fritillariae Cirrhosae and Radix Ampelopsis,respectively,contents of the diester-diterpenoid alkaloids are less than single Radix Aconiti Laterlis Preparata decoction. To study the contents of diester-diterpenoid alkaloids in decoctions with differents pH values water,the result was related to the pH values in decoctions,it is essential for diester-diterpenoid alkaloids contents. The HPLC and ESI-MS analytical results of the diester-diterpenoid alkaloids contents were accordant.A alkaloid of Radix Aconiti Kusnezoffii,was isolated from this plant,using a purification sheme of column chromatography based upon its polarity. Structural identification was performed using HPLC and ESI-MS analysis.This compound consists of dehydrated benzoylaconine is about 92%.Isolation technique of the compound was not reported erenow.Study on the biotransformation of dehydrated benzoylaconine of the metabolite of aconitine in rats intestinal bacteria by ESI-MS. dehydrated benzoylaconine was found to be converted by losses of CH3OH,CO and debenzoyltion in rats intestinal bacteriaThis thesis demonstrated that the decoction toxicity reduce when Radix Aconiti Laterlis Preparata combined with Rhizoma Pinelliae Preparata,Bulbus Fritillariae Cirrhosae,Radix Ampelopsis,and the others decoction toxicity increased. The experiment affirmed the theory of"Banloubeilianjigongwu"in point of chemical view.
