Objective:To explore the toxicity of Radix Aconite Laterlis when mix or decoct with Radix Glycyrrhizae,to investigate the effects of Radix Glycyrrhizae on toxicological pharmacokinetic parameter of Radix Aconite Laterlis,and discuss the alexipharmic effect of Radix Glycyrrhizae on Radix Aconite Laterlis.Methods:Note the mortality of mice,compare the date of Radix Aconite Laterlis with that of the mixture,investigate the influence of Radix Glycyrrhizae to the toxicity of Radix Aconite Laterlis.Draw the cardiograph of frog,determine the influence of Radix Glycyrrhizae on the toxicity of Radix Aconite Laterlis on heart.Determination of the ip LD50 and apparent pharmacokinetic parameters of Radix Aconite Laterlis and the mixture of Radix Glycyrrhizae and Radix Aconite Laterlis by acute moritality of mice, then analyse the process in the body.Results:Radix Glycyrrhizae can reduce the toxicity of Radix Aconite Laterlis in mice distinctly,especially when Radix Glycyrrhizae and Radix Aconite Laterlis in the proportion of 1:1.The toxicity of Radix Aconite Laterlis on heart reduce when decoct with Radix Glycyrrhizae.Decoct Radix Aconite Laterlis and Radix Glycyrrhizae together,can make the ip LD50 in mice improve from 5.27 to 9.36 g/kg.Determination of apparent pharmacokinetic parameters of Radix Aconite Laterlis and the mixture by acute moritality of mice,the result indicate that their biotransformation both belong to first-order elimination kinetics,one open compartment model.The apparent half life of elimination were estimated to be 1.18h and 0.92h.Conclution:Decoct Radix Aconite Laterlis together with Rxdix Glycyrrhizae can reduce the toxicity and improve the ip LD50 of Radix Aconite Laterlis in mice.The research of apparent pharmacokinetic parameters indicate that Radix Glycyrrhizae may change toxicological pharmacokinetic parameters of Radix Aconite Laterlis,shorten half life of elimination and increase apparent volume of distribution.
