| Objective:To study the mechanism of early high calcium intake to adulthood obesity through the high calcium intervention to ablactation rats and adult obesity induced by high fat diet. Methods:1. Fifty male Wistar rats were randomly divided into obesity-induced groups and normal control group, with high fat diet and basic diet for 8 weeks. The change of body fat was observed and at the end of 8 weeks, the levels of leptin and insulin were compared.2.120 male Wistar rats were divided randomly into normal control group, high dose calcium group, medium dose calcium group and low doses calcium group, with basic diet and high calcium diet for 4 weeks. After intervention, blood glucose, blood fat, serum calcium, the levels of leptin, insulin and PTH were compared. Hepars and kidneys for pathological studies were taken by HE dyeing. Subsequently, all rats were fed basic diet for 3 weeks and indexes above were observed, adipose tissue of high-testosterone weeks and kidney weeks were weighed for calculating the body fat content. Normal feed rats were randomly divided into normal controls and obesity induction group. Obesity induction group, high dose calcium group, medium dose calcium group and low doses calcium group were fed high-fat food. After 8 weeks rats were executed for testing the indexes above. And adipose tissue of high-testosterone weeks and kidney weeks were weighed for calculating the body fat content. RT-PCR was used for analysising the level of UCP2 gene expression. Results:1. The weights, serum TC, LDL-C, leptin and insulin levels of fat rats were significantly higher than normal control group.2. Before high-fat intervention, fat/body ratio of three high calcium groups were not higher than normal control group. Weight of low and medium dose calcium groups were not higher than normal controls. Weight of high dose calcium group were obviously lower than the other groups.3. Weight and weight growth of three high calcium groups were below obesity induction group; weight and fat/body ratio of three high calcium groups were no difference with normal control group; weight growth of low doses and high doses of high calcium groups were no difference with normal control group.4. After fatty intervention serum fasting blood sugar content did not differ.5. After fatty intervention serum triglyceride levels of high dose high calcium group were significantly lower than obesity induction group and normal controls consistently.6. After fatty intervention blood leptin and insulin level of three high calcium groups were significantly lower than obesity induction group. Parathyroid hormone level of three high calcium groups obviously rised compared with before high calcium intervention.7. Expression level of UCP2 gene of obesity induction group and high doses of high calcium group were obviously lower than normal controls, low dose and medium dose high calcium group. Expression level of UCP2 gene of medium dose high calcium group was significantly higher than normal control group. Conclusion:In rats'Early life high calcium intake can continue to affect adulthood obesity formation induced by high-fat feed, increase sensitivity to leptin, prevent resistance to insulin sensitivity, increase expression level of UCP2gene, make the body maintain normal body fat by intaking low dose and medium dose high calcium during the period of high-fat diet. |
