Experimental Study Of Acupuncture Chronic Fatigue Caused By Multiple Factors In Rat Hippocampal 5-ht ,5-ht1a Receptor Mrna Expression Intervention

Objective:To compare the two kinds of CFS models of rats,which are obtained by the methods of double stress factors and multiple stress factors.And to discuss the possible mechanism of acupuncture which effect in the hippocampus 5-HT,5-HT1ARmRNA of CFS model of rats by multiple stress factors.Method:It is through a multiple stress factors with constraint,swimming, day-night reversal and empty bottle stimulation to obtain the chronic fatigue model of rats.The model groups are exposed for 21days to different chronic stressors.Observing the behavior changes of the rats and examining the content of the CORT,ACTH in serum,making a distinction between the methods of double factors stress and multiple factors stress.Then the acupuncture group rats are acupunctured in bilateral Zusanli,Taichong,Baihuionce daily,and this group accepted multiple factors stress at the same time for 21days.And examining the content of 5-HT,5-HT1ARmRNA in the hippocampus.Results:In this experiment,the multiple factors model is more effective than double factors model in the aspect of the general conditions,the behavior changes,the content of the CORT,ACTH in serum.For example,the physical fatigue, the spatial learning and memory deficits of the multiple factors model rats are more serious than the double factors model rats,and the changes of emotion become more and more complicated and various,not only manifest depressive emotion, the content of the content of the CORT and ACTH in serum are even higher too. Through the treatment of acupuncture,the body weight and the body temperature of the acupun-cture group rats become much higher than model rats,the ability of spatial learning and memory,the physical fatigue and the changes of emotion become better too.And after acupuncture treating,the 5-HT,5-HT1ARmRNA in the hippocampus are higher than model group.Conclusion: 1.The behavior changes of chronic fatigue rat models induced by multiple factors are close to human CFS symptom,it can be used as an animal model to study CFS.2.The acupuncture not only can markedly reverse the physical fatigue and the mental fatigue of model rats,but also can improve the content of the 5-HT, 5-HT1AmRNA in the hippocampus.3.Regulating the expression of 5-HT1ARmRNA in the hippocampus by acupuncture may be one of the effective mechanisms in treating CFS.
