| Objective: Cortex Eucommiae (duzhong) is the dried bark of Eucommia ulmoides Oliv.(Eucommiaceae).The gum of duzhong's barks restricts the extraction of active components, so duzhong is always processed before used in clinic. However, no parameter of procedure and quality control method is regularized in Pharmacopoeia of the People's Republic of China. To resolve above problems, this thesis began with the study on the impact of procedure to chemical constituents, combining with the isolation of chemical ingredients, to definite the chemical compositions changing remarkably led by procedure. With above compounds as quality control marks the HPLC fingerprint was established for quality determination of duzhong. At last, according to the influence of procedure to active constituents, we optimized parameter of procedure to formulate the reasonable procedure of duzhong with saline water and established the quality control standard of processed product. The result provided safeguard for validity and security of clinical medication.Material and method:Material: The dried bark of Eucommia ulmoides Oliv. (Eucommiaceae) commerced from herbal store.Method: 1. The variation of chemical ingredients between crude drug and processed duzhong was analyzed and the impact of procedure to chemical constituents was studied by means of UV, TLC and HPLC.2. The chemical constituents changing significantly after processing were isolated and purified by various chromatographic methods including silica gel column chromatography, Sephadex LH-20 column chromatography and HPLC. Their structures were identified by the physic-chemical property and spectra data.3. The chemical ingredients changing remarkbly of duzhong after processing were identified by TLC and HPLC.4. Based on the principle of "breaking silk and retaining active constituents", the procedure parameter of duzhong was optimized and the quality standard of processed product was established. Result1. The impact of procedure to chemical compositions revealed that temperature and time played a significant role in duzhong's silk broken and chemical ingredients' changing. When processing temperature over 140℃, the silk could be broken and chemical compositions changed significantly.2. Sixteen compounds were isolated from duzhong. Eleven of them were isolated and purified from alcohol extractive of crud drug by various chromatographic methods. Gardendiol was isolated from duzhong for the first time. Five compounds were isolated and purified from alcohol extractive of processed duzhong.3. The content of most ingredients decreased after processing at high temperature. Genipin, gardendiol, eucommiol and geniposidic acid reduced remarkably. Pinoresinol diglucoside, syringaresinol diglucoside and medioresinol diglucoside were also lost. Pinoresinol, epipinoresinol and medioresinol increased after processing were the aglycones of lignan glycoside decreased obviously after processing from duzhong.4. Based on the principle of "breaking silk and not losing active components", the suitable procedure of duzhong was certained that Cortex Eucommiae was saturated with 2% saline water and heated at 140℃for 60 minutes. On the basis of the procedure the quality control standard of processed product was established.Conclusion:1. Based on the spectral-chromatographic assay, the impact of processing to chemical constituents was analyzed on macroscopic and microcosmic aspects for the first time in this thesis.2. For the first time, several chemical ingredients of duzhong changing remarkably after processing were ascertained. The HPLC fingerprint of duzhong was established with several active constituents as control marks. The result indicates that the impact of processing to chemical constituents is obvious, especially the content of lignans and iridoids reduces after processing at high temperature. In conclusion, all above results provide scientific evidence for the procedure of duzhong and the quality control standard of processed product.3. The procedure parameter of duzhong was optimized and the quality control standard of processed product was founded according to the processing principle of "breaking silk and retaining active components" firstly proposed by us in this thesis.4. The procedure we established in this paper is identical with the ancient documents of opinion that duzhong should be processed by "slow fire" and "pyretic stroke". |
