Mass spectrometry techniques, especially soft ionization mass spectrometry and itsassociated technique, have been proved to be one of the most powerful tools in the analysis oflarge bio-molecules and traditional Chinese medicines due to the high sensitivity, speed,resolution and low levels of sample consumption. High performance liquid chromatography(HPLC) techniques, comparing with other methods, have the advantages of high columnefficiency, sensitivity, separation speed. It has been applied widely in the analysis oftraditional Chinese medicine, not only used for quantitative determination, varietalsidentification, classification and separation, but also for the research of concoction, place oforigin, harvest-season and deposited condition. High performance liquid chromatographycoupled to electrospray ionization tandem mass spectrometry (HPLC-ESI-MS~n), whichcombines the ability of chromatographic separation and high exclusive properties of massspectrometry, has been used widely in identification of micro-impurity in drug degradationand bio-transform products of drug, pharmacokinetics, high-through put of combinedchemistry and analysis in natural products. Capillary zone electrophoresis method has theadvantage of separation speed, efficiency, low levels of reagent and sample consumption,long longevity capillary column, easy cleaning.It has widely used in component analysis oftraditional Chinese medicine.Firstly, compounds in Cortex Eucommiae have been quantitative analyzed by highperformance liquid chromatography (HPLC). The preparation technics of Cortex Eucommiaehas been optimized, and the optimal preparation conditions have been obtained. Furthermore,the chromatographic separation method of lignans in Cortex Eucommiae has been developed,in which 6 chromatographic peaks have been identified by high performance liquid chromatography coupled to electrospray ionization tandem mass spectrometry(HPLC-ESI-MS~n). As a result, the quality control methods of Cortex Eucommiae have beenestablished. The quantity change of main component showed that processing CortexEucommiae was very signifigative.Secondly, the content of main component chlorogenic acid in Cortex Eucommiae hasbeen determined by Capillary zone electrophoretic method first time. A new and efficientmethod has been provided for quality control of Cortex Eucommiae and medicine includingCortex Eucommiae.Finally, the changed regularity of components content in Cortex Eucommiae combinedwith Radix Dipsaci has been quantitative analyzed by HPLC. The experiment results providechemical basis for studying traditional Chinese medicine prescription.
