Projective: To observe the Jiannao soothe the nerves of acupuncture on post-stroke depression (Post stroke depression, PSD) rats before and after treatment study body weight and behavior and brain BDNF, dopamine 3 receptor (D3),βreceptor mRNA expression levels to explore Jiannao soothe the nerves of acupuncture treatment of post-stroke depression mechanism.Methods: 1. Divide the rats randomly into 6 groups as: blank array, model one, acupuncture one, fluoxetine one and acupuncture-fluoxetine one.2. Produce the animal model with disease-verification, i.e. the rat model with Post stroke depression (PSD) by combining the cerebral ischemia caused by removing blood vessel from the cerebral cortex with chronic unpredictable mild stress (CUMS) and separation.3. Measure the body weight, content of sugar water drunk, and score of the open-field test under the chronic unpredictable mild stress (CUMS) for each rat on the 1st day, the 8th, the 15th, the 22nd, and the 29th, observe the effect of the Strengthening-Brain-and-Tranquilizing- Mind Acupuncture on the body weight, consumption of the sugar water, and the score of open-field test.4. Experiment at the first 36 days all rats were sacrificed by reverse transcription- polymerase chain reaction (RT-PCR) determination of post-stroke depression in rat brain BDNF, dopamine 3 receptor (D3),βreceptor mRNA expression level observed Jiannao soothe the nerves of acupuncture on post-stroke depression rat brain BDNF, dopamine 3 receptor (D3),βreceptor mRNA affected.Results: 1.Observe the effect of the Strengthening-Brain-and-Tranquilizing-Mind Acupuncture on the body weight and the behavior of the rat with the post stroke depression (PSD): In comparing with the model array, the body weight of the acupuncture array, acupuncture-fluoxetine one and fluoxetine was increased, the consumption of the sugar water was increased, and the score of the open field test was higher. And there is no difference between the acupuncture array and fluoxetine one.2. The body weight of the rats in the model array is lighter than others. And the content of the cane sugar water drunk and the score of the movement levelly and vertically are all less than the blank array's. If increasing the body weight of the PSD rat there is no different effect between them (P>0.05) but the effect on the fluoxetine array started more quickly; the acupuncture can increase the consumption of the sugar water by the rat and the effect likes the fluoxetine (P>0.05); the acupuncture can increase the movement of the rat levelly and the effect starts more quickly, but the effect on the fluoxetine array is better (P<0.05); the acupuncture is able to increase the movement of the rat vertically; in contrast with other two arrays, the effect is the best one (P<0.05).3. Jiannao soothe the nerves of acupuncture on post-stroke depression in rat brain BDNFmRNA impact: with a blank group, model group, the expression of rat brain BDNFmRNA significantly reduced (P <0.01); Compared with model group, the group of large BDNFmRNA expression of rat brain tissue was significantly increased (P <0.01), one of the most obvious increase in fluoxetine group, followed by acupuncture and acupuncture + fluoxetine group; but acupuncture + fluoxetine group and acupuncture group compared to no significant difference (P> 0.05).4. Jiannao soothe the nerves of acupuncture on post-stroke depression in rat brain dopamine 3 receptor (D3) mRNA impact: model group, rat brain D3 receptor mRNA expression was significantly increased (P <0.01); with the model group ratio, acupuncture group and acupuncture + fluoxetine group of rat brain D3 receptor mRNA expression was significantly lower (P <0.05 or P <0.01), one of the most obvious lower acupuncture group; but fluoxetine group and model group compared to no significant improvement.5. Jiannao soothe the nerves of acupuncture on post-stroke depression in rat brain the effects ofβ-adrenergic receptor mRNA: with a blank group, model group, rat brain tissue on the Su-renalβ-adrenergic receptor mRNA expression can be significantly higher (P <0.01 ); Compared with model group, acupuncture group and acupuncture + fluoxetine group on rat brain kidney Su-toβ-adrenergic receptor mRNA expression levels decreased significantly (P <0.05 or P <0.01), one of acupuncture group reduced the most obvious; fluoxetine group compared with model group, no significant difference (P> 0.05).Conclusion: 1. By using the complex modeling method, combining the cerebral ischemia caused by removing blood vessel from the cerebral cortex with chronic unpredictable mild stress (CUMS) and the separation, the animal model with the post stroke depression (PSD) can be produced successively.2. Determination of the indicators in this study showed: Jiannao Anshen acupuncture can regulate body weight, behavior study, dessert consumption, increase the level of BDNFmRNA to regulate the D3 receptor mRNA level and regulation of rat brain on the Su-kidney canβ-adrenergic receptor mRNA level to improve post-stroke depression symptoms of depression in rats, and its considerable efficacy and fluoxetine.3. The Strengthening-Brain-and-Tranquilizing-Mind Acupuncture is an effective method to treat the post stroke depression (PSD) and it is worth to be researched and developed further.
