Divide the rats randomly into 6 groups as: blank array, model one, acupuncture one, fluoxetine one and acupuncture-fluoxetine one.2. Produce the animal model with disease-verification, i.e. the rat model with Post stroke depression (PSD) by combining the cerebral ischemia caused by removing blood vessel from the cerebral cortex with chronic unpredictable mild stress (CUMS) and separation.3. Observe the effect of the Strengthening-Brain-and-Tranquilizing-Mind Acupuncture on the body weight and the consumption of the sugar water of the rat with the post stroke depression (PSD): In comparing with the blank array, the body weight and the consumption of the sugar water of the model array was decreased; In comparing with the model array, the body weight of the acupuncture array, acupuncture-fluoxetine one and fluoxetine was increased, the consumption of the sugar water was increased, And there is no difference between the acupuncture array and fluoxetine one. and the score of the open field test was higher.2. Observe the effect of the Strengthening-Brain-and-Tranquilizing-Mind Acupuncture on behavior of the rat with the post stroke depression (PSD): The score of the movement levelly and vertically are all less than the blank array's. The acupuncture array can increase the score of the movement levelly of the PSD rat quickly, but the effect on the fluoxetine array was the best one; The acupuncture array can increase the score of the movement vertically of the PSD rat, and the effect was better than the others.3. Jiannao soothe the nerves of acupuncture on post-stroke depression model rat 5-HT1A ,5-HT2A, receptor mRNA level: the treatment of PSD rats through 5-HT1A receptor expression ,5-HT2A receptor expression increased. And fluoxetine, through increase of 5-HT1A receptor expression, reduced 5-HT2A receptor expression in the treatment of PSD are different, acupuncture, acupuncture plus fluoxetine are raised through 5-HT1A receptor expression, reduced by 5-HT2A body,βreceptor, D3 receptor expression play a role in the treatment of PSD. The results show that fluoxetine mainly through the effects of 5-HT receptor expression,not only acupuncture, acupuncture plus fluoxetine in addition to the effects of 5-HT receptor expression, but also can adjust the NE receptors, dopamine receptors, thereby played many parts of many therapeutic targets.Conclusion: 1. By using the complex modeling method, combining the cerebral ischemia caused by removing blood vessel from the cerebral cortex with chronic unpredictable mild stress (CUMS) and the separation, the animal model with the post stroke depression (PSD) can be produced successively.2. Determination of the indicators in this study showed: Jiannao Anshen acupuncture can affect the 5 - hydroxytryptamine receptor 1A (5-HT1A), 5 - hydroxytryptamine 2A (5-HT2A) mRNA expression levels.3. The Strengthening-Brain-and-Tranquilizing-Mind Acupuncture is an effective method to treat the post stroke depression (PSD) and it is worth to be researched and developed further. |
