Objective:Treatment of Mongolian medicine acupuncture is applied on the forehead acupoint,lumen acupoint,shangyu acupoint and dinghui acupoint of rats with post-stroke depression,observing and analyzing the behavior,neurological deficits and effects on serum cytokines IL-1β IL-6,TNF-α of rats,identifying the effects and possible function of Mongolian medicine acupuncture treated on post-stroke depression.Method:SD rats were adaptively reared for one week and were randomly divided into four groups according to body weight:blank control group,model group,acupuncture group and drug group.There were 12 rats in the blank control group and 36 in each of the remaining three groups.After one week of adaptive feeding,except for the blank control group,the other three groups were prepared with a modified cerebral artery ischemia-i eperfiision model(MCAOIR),and reperfusion was performed 90 minutes after cerebral ischemia.Neurological deficit were scored 2h after awake in rats,and the Bederson score(1 score to 3 score)showed successful cerebral ischemia models then seven chronic unpredictable stress methods(CUMS)were used to combine the alone feeding to prepare a depression model,in which the blank control group was noirnal feeding without any treatment.Rat body weight was observed during the experiment,and the syrup experiment and open box experiment were performed after the end of chronic stress(28 days).Treatment method:The acupuncture group selects the forehead acupoint,lumen acupoint,shangyu acupoint and dinghui acupoint for treatment once daily,total for 28 days.The drug group was treated with fluoxetine hvdrochloride 4.06 mg/kg.d,gavage therapy for 28 days.The model group was given only distilled water and was intragastrically administered at 1 ml/100 g.The blank control group was normal feeding without any treatment.The syrup experiment and the open box experiment were carried out on the first day of treatment,the 14th day of treatment,and the 28th day of treatment.After the behavioral obsen ation,the animals were anesthetized,and conducted laparotomy,the abdominal aorta was taken for blood.Centrifugation at 3000 r/min for 15 min after clotting,then stored the serum for testing.The levels of serum IL-1β,IL-6 and TNF-αcytokines were detected by ELISA.All data were expressed by ’x=s,and P<0.05 was considered statistically significant.Results:1.The neurological deficit scores were obtained 2h after awake in rats,the Bederson scores of the rats in each group were(1 to 3).After 24h,the neurological deficit of the rats returned to normal,and the Bederson score was 0,indicating that the rats had mild dysfunction of the contralateral limbs and obvious neurological deficits after awake.After the end of chronic stress(28 days),the body weight and behavioral evaluation of the blank control group and the model group were compared,and the weight and behavioral evaluation results of the model group were significantly lower(P<0.05),indicating the PSD model was successfully prepared.Compared with the model group,the body weight,syrup consumption and vertical exercise level of the acupuncture group and the drug group increased significantly(P<0.05).There was no significant difteronce(P>0.05)in the acupuncture group and the drug group with body weight,sugar consumption and vertical movement level of rats.2.Comparison of seru IL-1β,IL-6 and TNF-α cytokines in the rats after treatment showed that:Compared with the blank control group,the serun levels of IL-1β,IL-6 and TNF-α in the model group were significantly increased(P<0.05):Compared with the model group,serum IL-1β,IL-6 and TNF-α cytokines in the acupuncture group and the drug group were siginiticantly decreased(P<0.05);There was no significant difference in serum IL-1β,IL-6 and TNF-α cytokines between the acupuncture group and the drug group(P>0.05).Conclusion:An animal model of post-stroke depression was succes sfully prepared by a rat model of middle cerebral artery occlusion reperfusion(MCAO/R)combined with chronic unpredictable stress stimulation(GUMS)and alone feeding.Acupuncture at the forehead acupoint,lumen acupoint,shangyu acupoint and dinghui acupoint is effective in treating post-stroke depression which can increase body woighth consumption of sugar and water,and increase the number of vertical movements,which can effectively reduce the levels of serum IL-1β,IL-6 and TNF-αcytokines in rats,and there is no significant difference in the efficacy of fluoxetine hydrochloride in the treatment ot post-stroke depression.This experimental study confirmed that the therapeutic eftect of Mongolian acupuncture is similar to that ot fluoxetine hydrochloride,and as a traditional treatment of Mongolian medicine,it has fewer side effects and further research value and development.
