ObjectivesCompares through experiment pharmacology by the pharmacodynamics difference between decocting the behalf of prescription of Exterior-releasing Methods and Promoting Blood Circulation and Removing Blood Stasis methods ,GQD(Ganmao Decoction) and LMD (Lemai Decoction),which are prepared by machine-boiling method and traditional earthenware pot-boiling method.Methods1 Social researchThrough to 304 patient's investigations, understanding the community's attitude an the reasons for thea attitude to traditional Chinese medicine compound decoction method.2 Experimental study2.1 Establish the dry yeast fever in rats by subcutaneous injection suspension model,fill the rats with GQD liquid separately decocted by Conventional traditional earthenware pot-boiling method and medicinal herbs machine according to the standard decoction method as well as gives the aspirin decoction ,then Observation of changes in rectal temperature in rats,determinat the two methods decocted GQD liquid's influence on the the rats blood serum IL-1βand the TNF-a content, by enzyme-linked immunosorbent assay methods, determinat the two methods decocted GQD liquid's influence on the the rats blood serum No content, by Colorimetry methods.2.2 Establish the hyperlipidemia rat model and applicat intraperitoneal injection of isoproterenol to establish myocardial ischemia composite rat model,and establish simple cardiac muscle ischemia rat model.Observes the effects on model rats' electrocardiogram Q wave, ST segment and heart rate realated to LMD liquid decocted by two decocting methods, As well as the effects on the content of rat's The Creatine kinase (CK), the phosphoric acid to stir up the sour isozyme (CK-MB) and the rats total cholesterol (HODL), the triglyceride (TG), the high density lipoprotein (HDL-C).Results1 Questionnaire survey resultBy the three age groups of a total of 304 people chosen decocting surveys, The results showed that 52.7 percent in the number of people mainly used earthenware cooking pot decoct traditional Chinese medicine in recent years,40.1 percent in the number of people mainly used decocts medicinal herbs machine cooking pot decoct traditional Chinese medicine in recent years. Ruleing out the possibility of objective factors, purely looking from the curative effect, support Thought that the curative effect of traditional earthenware pot-boiling method is better are to account for 78.6%,while support machine-boiling are account for 6.6%.2 The Pharmacology quite studies results between the decoctions of GQD prepared by machine-boiling method and that preparde by earthenware pot-boiling methodThis experiment was altogether repeated 3 times, each rat in each group were 10. About the third batch experiment ,giving dry yeast suspension hypodermic injection 4h later, The rectal temperature of MD(model group) rats was (37.6±0.26)℃,8h later, The rectal temperature of MD achieves the peak value which was (39.46±0.29)℃;12h later, the rectal temperature of MD receds to(38.34±0.4)℃.In the third batch experiment,the GMG (decocting machine group)decoction decocted by traditional earthenware pot-boiling method curative effect acts only after being given 1hour later, The rectal temperature of ECPG(earthenware cooking pot group) rats was (37.13±0.16)℃, the change rate was( 0.24±0.13)%, has non-significance the difference with the NG (normal group); The rectal temperature of DMG (decocting machine group) rats was( 37.4±0.13)℃, there was a significant difference with ECPG's,p<0.05. After making the mold 8h or given medicine 7h later, The rectal temperature of ECPG rats was (38.62±0.17)℃, the change rate was (1.74±0.15)%,was lower than both MG and DMG, the difference was significant,p<0.05.In the other two batch experiments,the time spot which the ECPG's rectal temperature significantly lower than GMG 's and MG's were separately in gavage 5h ,9h later and 3h,7h,9h later. The experimental result can partly repete the 3rd experimental result,and there were no opposite situation appear.In the third batch experiment, after making the mold 8h or given medicine 7h later, IL-1βin rats shows that the ECPG's IL-1βcontent was no difference with the NG's and significantly lower than GMG's, the ECPG's IL-1βcontent was(22.86±8.49 )pg/ml,the GMG's was (41.44±9.3)pg/ml,there was significant difference between them, p<0.05。3 The Pharmacology quite studies results between the decoctions of LD prepared by machine-boiling method3.1 ECG comparative analysis resultsQ-wave changes in rats ECG After injecting Iso 5min and 20min ,the Q-wave changes of ECPG' rats were obviously lower than MGⅡand MGⅠ, with the remarkable difference in statistics,p<0.05; injecting Iso 20min later,the GMG' Q wave change(-0.13±0.03)mv has no remarkable difference with MGⅡ(-0.09±0.04)mv; injecting Iso 20min later, there was remarkable difference between MGⅡ(-0.13±0.03)mv and MGⅠ, p<0.05. injecting Iso 24hours later, the Q-wave amplitude changes of ECPG's has no difference with its normal value before making the mold,while the GMG and two model groups were obviously both higher than their normal value before making the mold, p<0.05.ST section change in rat ECG after injecting Iso 20min,the elevation range of ST section of ECPG was (0.04±0.02)mv which was Significantly lower than that of GMG(0.08±0.03)mv, with statistically significant differences, p<0.05;the ECPG was significantly lower than both MGⅡand MGⅠafter injecting Iso 5min,20min,and 24 hours, p<0.05; The change scope result of ST section of GMG has no difference with MGⅠafter injecting Iso 20min. Compared MGⅠwith MGⅡ, MGⅡwas Significantly higher than MGⅠafter injecting Iso 5min and 20min,with a statistically significant difference,p<0.05。The occurrence of Premature ventricular contractions(PVC) in rats ECG after injecting Iso After injecting Iso 5 min,the ECPG's occurrence of PVC was (1.43±1.62)times which was lower both than MGⅠ(4.86±2.04)times and MGⅡ(8±3.65)tims , with a statistically significant difference ,p<0.006. During the time 5min and 10min after injecting Iso, the ECPG's occurrence of PVC was(1.43±1.62)times and GMG's was(3.57±1.62)tims,they both were lower than MGⅠand MGⅡ, (8.71±1.38)and(12.14±3.63)tims, with a statistically significant difference,p<0.006. During the time 10min and 30min after injecting Iso, the ECPG's occurrence of PVC was (3.71±3.45)times which was lower both than MGⅠ(11.14±3.13)times and MGⅡ(20.86±7.73)tims , with a statistically significant difference ,p<0.006; the GMG's occurrence of PVC was(5.14±2.19)times,was only significantly lower than that of MGⅡ.There was no no significant difference between ECPG and GMG in each time.3.2 Enzymogram analysis resultsTakes the blood of rats after after injecting Iso 24 hours,and examinate the enzymogram. the CK content of ECPG was(121±24.77)IU/l, the CK-MB content of ECPG was(239.43±29.88)IU/l, the CK content of GMG was (170.57±40.09 )IU/1, its CK-MB content was (305.29±78.67)IU/l,there was no difference both in CK an CK-MB between the two group,and they were both lower than the two model groupⅠandⅡ, with a statistically significant difference, p<0.05. the CK content of MGⅠwas(323.14±138.58)IU/l and CK-MB content was (447.14±159.47)IU/1; the CK content of MGⅡwas (491.43±163.16)IU/1 and CK-MB content was (617.71±122.93)IU/1, MGⅡwas significantly higher than MGⅠboth CK and CK-MB, p<0.05.3.3 Blood lipidFeeding the rats with High-fat forage for 10 days, the ECPG 'content of HODL,TG and HDL-C were (1.68±0.27)mmol/l,(0.64±0.29)mmol/l and (1.09±0.16)mmol/l, the GMG 'content of HODL,TG and HDL-C were(1.98±0.20)mmol/l,(0.56±0.2)mmol/l,(1.18±0.07)mmol/l,there was no significant difference between ECPG and GMG. The ECPG's CHOL,TG,HDL-C contents were all with no significant difference with the contens before being fed with High-fat forage,(1.6± 0.30)mmol/l,(1.02±0.45)mmol/l and (1.05±0.21)mmol/l.while the GMG's CHOL content (1.49±0.13mmol/l )and HDL-C(1.0±0.14mmol/l )were both significantly higher than he contens before being fed with High-fat forage, (1.49±0.13mmol/l) and(1.0±0.14mmol/l ),p<0.05,the TG content of GMG was lower than the content before being fed with High-fat forage, with a statistically significant difference,p<0.05.Result(1) There is significant difference in curing yeast-induced fever in rats between the GMD decoction decocted with machine-boiling method and traditional earthenware pot-boiling method.The GMD decoction decocted with traditional earthenware pot-boiling method is obviously better than that decocted by decocts medicinal herbs machine in temperature decrease effect and the speed of affect.(2) In regard to the effction on inflammatory cytokines, experimental results shows that:GMD decoction decocted by traditional earthenware pot can Obviously inhibit the Serum IL-1βof rats to raise.(3) There are some significant difference in curing Iso-induced myocardial ischemiain rats between the LMD decoction decocted with machine-boiling method and traditional earthenware pot-boiling method, at the time 20 minutes after the injecting the Iso, the LMD decoction decocted earthenware pot is obviously better than the decoction decocted by decocts medicinal herbs machine in improveing myocardial ischemiain and the role of reducing ST-elevated,the other time point,though there is difference, however, is no statistically significant.(4) In regard to the effction on enzymogram and blood lipids,there is no statistically significant between the two decocting methods.(5) It is proved from enzymes and ECG that the extent of myocardial injury caused by the Iso-induced myocardial ischemiain Composite rat model which based on hyperlipidemia rat model is serious than Iso-induced myocardial ischemiain rat model, Inject Iso on hyperlipidemia rat is easier to establish myocardial ischemiain model.(6) Compare the pharmacodynamic experiments with GMD and LMD decoction decocted by machine-boiling standard method to traditional earthenware pot-boiling standard method,the Pharmacodynamics of decoction with traditional earthenware pot-boiling method has advantages over decoction with machine-boiling standard method in inhibiting temperature rise and curing rat'myocardial ischemia.
