Analysis And Comparison Of The Extracting Machine And Traditional Casserole Medicine Or The Legal System Preparation Of Bazhen Yimu Side Decoction | Posted on:2010-09-03 | Degree:Master | Type:Thesis | Country:China | Candidate:C Feng | Full Text:PDF | GTID:2204360272994860 | Subject:Chinese medicine | Abstract/Summary: | PDF Full Text Request | ObjectiveIn this paper,we produce Bazhenyimu Decoction by decocting machine and traditional casserole decoction,then compare the two kinds of decoction by determination of the amounts of extract and active ingredients.In the end,we come to the conclusion of the impact on Bazhenyimu Decoction of the two decoction methods.MethodsIn this paper,Bazhenyimu Decoctions were produced by decocting machine and traditional casserole decoction in accordance with the present decoction standards of both traditional casseroles decoction and decocting machine.Then the total amounts of extract as a overall indicator and the contents of paeoniflorin,ferulic acid and stachydrine as active ingredients indicator of the two kinds of Bazhenyimu Decoctions were determined.In the end,a conclusion of the impacts of the two kinds of decoction methods on Bazhenyimu Decoction in chemistry was conducted by comprehensive analysis of results.In this paper,the experiments consist of four parts:1 The processes of the two kinds of decoction of Bazhenyimu Decoction were optimized.Bazhenyimu Decoctions were produced by both decocting machine and traditional casserole decoction in accordance with the optimized processes,and then the two kinds of Bazhenyimu Decoction were compared on characteristics and volume.2 The total amounts of the two kinds of Bazhenyimu decoction were determined based on the method of determination of water extract in "Chinese Pharmacopoeia".3 A quantitative HPLC-DAD method was established to determine the contents of paeoniflorin and ferulic acid simultaneously in Bazhenyimu Decoction.The contents of Paeoniflorin and ferulic acid were determined in Bazhenyimu Decoction, Paeonia lactiflora,Ligusticum chuanxiong Hort and Angelica sinensis.The extract rates of paeoniflorin and ferulic acid from the herbal pieces to Bazhenyimu Decoction were calculated.In the end,the contents of paeoniflorin and ferulic acid of the two kinds of decoction were compared.4 A quantitative HPLC-ELSD method was established to determine the contents of stachydrine in Bazhenyimu Decoction.The contents of stachydrine were determined in Bazhenyimu Decoction and Leonurus japonicus Houtt.The extract rates of stachydrine from the herbal pieces to Bazhenyimu Decoction were calculated. In the end,the contents of stachydrine of the two kinds of decoction were compared.Results1 Decoction characteristics:The Bazhenyimu Decoction produced by the traditional casserole decoction was deeper in color,thicker in smell and stickier than that produced by decocting machine.2 Decoction volume:the volume of Bazhenyimu Decoction produced by the traditional casserole decoction was significantly lower than that produced by decocting machine(p<0.05).3 The amounts of extract:the amounts of extract of Bazhenyimu Decoction produced by the traditional casserole decocting and machine decocting had no significant difference(p>0.05).4 A quantitative HPLC-DAD method was established to determine the contents of paeoniflorin and ferulic acid simultaneously in Bazhenyimu Decoction.The calibration curve are:paeoniflorin(230nm):Y1=1614355X1-41391.2,r=0.99979,and the calibration curve is linear at ranges of 0.07904~1.976μg;ferulic acid(322nm): Y2=6196894X2-57151.2,r=0.99990,and the calibration curve is linear at ranges of 0.02038~2.038μg.Average recoveries are:Paeoniflorin 99.45%,RSD=4.08%(n=6); ferulic acid 100.67%,RSD=3.61%(n=6).The contents of paeoniflorin and ferulic acid in Bazhenyimu Decoction produced by the traditional casserole decoction and decocting machine had no significant differences(p>0.05).5 A quantitative HPLC-ELSD method was established to determine the contents of stachydrine in Bazhenyimu Decoction.The calibration curve is:lnY=1.283145lnX +13.18717,r=0.9996 and the calibration curve is linear at ranges of 0.249~9.96μg. The average recovery rate was 96.90%,RSD=1.73%(n=6).The contents of stachydrine in Bazhenyimu Decoction produced by the traditional casserole decoction and decocting machine had no significant difference(p>0.05).ConclusionIn a word,although the Bazhenyimu Decoction produced by the traditional casserole decoction and decocting machine have significant differences on characteristics and volume(p<0.05),the amounts of extract as a total indicator and the contents of the active ingredients indicators including paeoniflorin,ferulic acid and stachydrine of the two kinds of decoction have no significant differences(p>0.05).
