Background and Objective:The methylation status change of the epigenetic modification is a key factor in the carcinogenic effect,the hypermethylation of CpG island in the promoter is the inactivation of tumor suppressor gene leading to a major mechanism.Environmental changes apparent change in gene research of epigenetic modification is very hot and the choice of the paper material was the plant estrogen environment quercetin,researching on colon cancer cell lines RKO growth,cell cycle and apoptosis in through the detection of tumor suppressor gene p16 activated of high methylation status changes,to explore its inhibitory mechanisms.Methods:We selected human colon cancer cell line RKO for the study,of which the p16 CpG island in the promoter is hypermethylation.The growth inhibition was assayed by MTT; Cell cycle and apoptosis of RKO cells were observed by flow cytometry technique; application of reverse transcription-polymerase chain reaction(RT-PCR) detected p21,p27 and p16 gene expression of mRNA and the application of Western blot(Western blot) detected p21,p27,p53 and p16 expression changes.Human colorectal cancer cell line RKO was treated in the presence of different concentrations of quercetin(5,10,20, 50μmol/L) for 5 days.The methylation status of promoter in p16 gene was analysed by MSP(methylation-specific PCR).Results:MTT assay showed that quercetin significantly inhibited cell growth and had dose-dependent effect;Flow cytometry technique showed that with 5μmol/L quercetin treatment cell cycle was arrested in G0/G1 phase;apoptosis rates of 5,10,20μmol/L groups were 23.4%,24.2%,47.9%and apoptosis rate of the control group was 13.2%. mRNA and protein expression levels of p21,p27 and p16 were increased,the protein expression levels of p21,p27,p53 and p16 were increased.The hypermethylation of p16 gene had been successfully reversed aider 120h disposal of quercetin. Conclusion:This paper shows that quercetin has significant growth inhibition on RKO cells. Quercetin has an effect on cell cycle and cell apoptosis.The changes of the tumor suppressor gene p16 promoter methylation status prompted quercetin may be play a role in anti-tumor through the mechanism of epigenetic modification for the resumption of tumor suppressor gene expression.
