| There are rich in nutrients in tartary buckwheat, including flavonoids, highly reactive protein, amino acids, microelements etc, which could reduce blood pressure, resist caducity and improve diabetic etc. Tartary buckwheat has been received more and more people’s favor as a kind of health food. And flavonoids from buckwheat, as an extremely nutritious substance, exerts a variety of biological activities, such as anti-oxidative, anti-mutagenic, free radical scavenging, anti-virus etc. In recent years, its unique anti-cancer function is concerned greatly. In this paper, the flavonoids quercetin and Isoquereetin extracted from buckwheat as experimental materials were deeply researched their antitumor effect.We studied the antitumor effects of quercetin and Isoquereetin from tartary buckwheat by treating SGC-7901cells. MTT assay showed that quercetin and Isoquereetin could inhibit the growth of SGC-7901cells in time and concentration dependent manners, the inhibitory effect of quercetin is stronger than Isoquereetin. DAPI nuclear staining showed that nuclear condensed, chromosome condensation and apoptotic bodies produced by treating SGC-7901cells with quercetin and Isoquereetin. Cell injury healing experiment indicated that quercetin and Isoquereetin could suppress cells migration in certain concentration range. And the inhibiting migration capacity of quercetin was bigger than that of Isoquereetin.The SGC-7901cells were treated with quercetin and Isoquereetin, results of flow cytometry showed that the cells of G1phase and S phase decreased, a lot of cells arrested in G2/M phase and the ratio of apoptosis cells increased, apoptosis-inducing effect of quercetin is more obviously. The SOD results showed quercetin and Isoquereetin can be elevated SOD activity, the antioxidant activity of quercetin is strongerer than that of Isoquereetin. SGC-7901cells were treated with quercetin, the results of RT-PCR showed that the transcription level of P53, P21increased and CyclinD1, CDK2, CDK7decreased. SGC-7901cells were treated with quercetin and Isoquercetin, the results of Western blot showed that the protein expression level of P21, P53were increased, the protein expression level of CyclinDl was decreased.The signal transduction pathways of cells apoptosis are very complex and extremely subtle, it needs many signal molecules to participate. The molecular mechanism of flavonoids inducing cells apoptosis still need to be further studied, and the thesis laid a experimental basis for this complex mechanism and the exploitation of the value of buckwheat as drugs, the study is originality and meaningful. |
