Objective:To investigate the expression and clinical significance of 4-1BBL in the tissue of cervical intraepithelial neoplasm and cervical cancer ,in order to further explore the role of 4-1BBL in carcinogenesis of cervical carcinoma which can provide the theory according to early prevention and treatment for cervical cancer.Methods: The mRNA expression of 4-1BBL was evaluated by means of RT-PCR from 58 cases of cervica1 cancer,25 cases of cervical intraepithelia1 neoplasmâ…¢(CINâ…¢). Meanwhile, the expression of the protein level of the 4-1BBL was detected by using SP immunohistochemical methods on paraffin-embedded sections from the corresponding positive specimen.Result: (1)The PCR assay shows that there was a strap in the 574bp area which indicate that 4-1BBL could be expressed in the tissue of cervical intraepithelial neoplasm and the cervical cancer.(2)The positive rate of 4-1BBL expression in CINIII,cervical cancer I and cervical cancer II are 24.0% ,14.7% ,8.7% respectively (3)The percentage of 4-1BBL-positive in the cervical cancer is lower than that in the cervical intraepithelial neoplasm ,however , there is no significant variability between them (P>0.05) (4) By Immunohistochemistry,the positive reaction mainly appeared in the membrane , as the damage is aggravated , the positive reaction is weaken.Conclusion: The lower expression rate of 4-1BBL may influence the anti-tumor immune response and contribute to carcinogenesis during the transformation of CIN into cervical cancer through immune escape which imply that it may be a target gene in immunogene therapy of cervical cancer ,and there is some objective clinical significance in it.
