Objective:Through the establishment of animal models and cytopathic effect model,we compare medicinal efficacy and potency between the geoherbs and non geoherbs Scutellaria baicalensis.This study establishes the scientific experimental basis for the clinical application.Method:Firstly,we establish the acute chemical liver injury models through intraperitoneal injection of carbon tetrachlorid to ICR mice,and detect some indexes such as ALT,AST,MDA,SOD.Secondly, we give cyclophosphamide to ICR mice to establishing immunosuppression models,then observe the protective effect of Scutellaria baicalensis to these mice and detect the effect of herbs on immune function indexes such as IgM,IgG and T lympholeukocyte subgroup.Thirdly,Staphylococcus aureus, Pseudomonas aeruginosa were used to antibacterial experiment in vitro and detected the the minimum inhibitory concentration (MIC)of different place of origin Scutellaria,meanwhile, coxsackievirus B3 and respiratory syncytial virus were used to anti-virus study,we compare the role of prevention and neutralization between the geoherbs and non geoherbs Scutellaria baicalensis and determine the therapeutic index(TI).Results:1.the effect on acute chemical liver injury mouse:The effect on ALT of the geoherbs and non geoherbs Scutellaria baicalensis were compared and the differences of them had statistical significance(P<0.01 or P<0.05) except between C3 and A3.The effect on AST were compared and the differences had statistical significance(P<0.01).The effect on ATPase activity in mouse livers were compared and differences had statistical significance(P<0.05) except between C2 and L2,C3 and L3 as well as A3. The effect on T-AOC and SOD were compared and the differences had statistical significance(P<0.01 or P<0.05) except between C3 and L3 as well as A3. The effect on MDA were compared and the differences had statistical significance(P<0.01 or P<0.05) except between C3 and L3.The effect on GSH were compared and the differences had no statistical significance(P>0.05) except between C2 and L2(P<0.01).The effect on GSH-Px were compared and the differences had statistical significance(P<0.05). 2.The effect on immunity suppression: The effect on IFN-γbetween the geoherbs and non geoherbs Scutellaria baicalensis were compared and the differences of high dose group had statistical significance(P<0.01 or P<0.05).The effect on TNF-αwere compared and the differences had no statistical significance(P>0.05) except between C1 and A1,C2 and A2 (P< 0.05). The effect on IgM were compared and the differences had statistical significance(P<0.05) except between C2 and A2 (P>0.05).The effect on IL-2 were compared and the differences had statistical significance(P<0.05) except between C3 and A3(P>0.05).3.Antibacterial and antiviral experiments: The TI of geoherbs Scutellaria baicalensis was 23.8 mg/mL and non geoherbs Scutellaria baicalensis were 16.9 mg/mL as well as 18.2 mg/mL in RSV group. The TI of geoherbs was 13.5 mg/mL and non geoherbs were 8.12 mg/mL and 7.28 mg/mL in CVB3 group.The MIC of geoherbs Scutellaria baicalensis on Staphylococcus aureus was 1.65 mg/mL and the non geoherbs Scutellaria baicalensis were 3.13 mg/mL and 3.13 mg/mL . The MIC of geoherbs Scutellaria baicalensis on Pseudomonas aeruginosa was 12.5 mg/mL, and the non geoherbs were 6.25 mg/mL and >25.0 mg/mL.Conclusions:Both the geoherbs and non geoherbs Scutellaria baicalensis have effects of immune regulation,sublimating antioxidant capacility of organism and protecting liver to a certain extent,but the efficacy of the geoherbs Scutellaria baicalensis surpass the non geoherbs.Meanwhile,the anti-bacterial and anti-viral potency of the geoherbs Scutellaria baicalensis overweigh the non geoherbs too.
